摘要
目的:探讨芦丁联合奥沙利铂对人胃癌SGC-7901细胞增殖及凋亡的影响。方法:采取对数生长期的人胃癌SGC-7901细胞作为研究对象,应用MTT法检测药物对胃癌SGC-7901细胞生长抑制率;Hoechst33258核染色观察细胞形态学变化;流式细胞术检测细胞周期及细胞凋亡率;Western-blot方法分析SGC-7901细胞中caspase-3、Bax和Bcl-2蛋白表达。结果:芦丁(100、200、300μmol/L)、奥沙利铂均可抑制SGC-7901的增殖,且呈剂量相关性;芦丁联合奥沙利铂与相同浓度奥沙利铂单用处理SGC-7901细胞相比,前者对SGC-7901细胞的抑制作用更显著(P<0.05);不同浓度芦丁与奥沙利铂联合处理SGC-7901细胞与奥沙利铂单用相比,前者显著提高了SGC-7901细胞的凋亡率(P<0.05)。芦丁作用后,caspase-3蛋白表达明显升高,Bcl-2/Bax蛋白表达的比值则明显降低(P<0.05)。结论:芦丁和奥沙利铂均可抑制SGC-7901细胞增殖并诱导其凋亡,其机制可能与上调caspase-3、Bax及下调Bcl-2等凋亡相关蛋白表达水平有关;两者联合应用具有一定的协同效应。
AIM： To study the effect of rutin combined with oxaliplatin on the proliferation and apoptosis of human gastric cancer SGC-7901 cells. METHODS： Human gastric cancer SGC-7901 cells in logarithmic growth phase were selected as the research object. Hoechst33258 nuclear staining was used to observe the morphological changes. Cell cycle and cell apoptosis were detected by flow cytometry. The expression of caspase-3, Bax and Bcl-2 protein in SGC-7901 cells were analyzed by Western-blot. RESULTS ： Rutin （ 100,200,300 μmol/ L） and oxaliplatin inhibited the proliferation of SGC- 7901 in a dose-dependent manner. Rutin combined with oxaliplatin, compared with the same concentration of oxaliplatin SGC-7901 cells, was significantly more effective on SGC-7901 cells （ P 〈 0.05 ）.Compared with oxaliplatin, SGC-7901 ceils treated with different concentrations of rutin and oxaliplatin significantly increased the apoptosis rate of SGC- 7901 ceils （P 〈 0.05 ）. The ratio of bcl-2/bax protein expression was significantly decreased （P 〈 0.05）. CONCLUSION ： Both curcumin and oxaliplatin can inhibit the proliferation of gastric carcer cells SGC-7901 and induce apoptosis. The mechanism may be related to up-regulation of caspase-3 and down-regulation of apoptosis-related protein such as bcl-2/bax. And the joint application of rutin and oxaliplatin presents some synergistic effect.
