摘要
目的 :探讨分析金钗石斛多糖对2型糖尿病大鼠心功能、炎症因子以及心脏组织中RIP3的表达特点,以为金叉石斛多糖的开发和2型糖尿病心肌损伤的治疗提供参考。方法 :60只SD大鼠随机分为正常组、模型组、金钗石斛多糖低、中和高剂量干预组以及阳性药物贝那普利治疗组,每组10只,除正常组外,其余5组给予高脂高糖饮食6个月构建2型糖尿病大鼠模型,模型建立成功后,正常组和模型组给予1 m L·kg^(-1)·d^(-1)生理盐水灌胃,金钗石斛多糖3个干预组分别给予金钗石斛多糖100、200、400 mg·kg^(-1)·d^(-1)治疗,贝那普利治疗组则给予贝那普利1 mg·kg^(-1)·d^(-1)灌胃治疗,治疗8周后处死大鼠,HE染色检测肝脏病理学,试剂盒检测心功能和炎性因子,实时定量RT-PCR和Western blot检测RIP3 m RNA和蛋白的表达水平。结果 :正常组大鼠毛色光滑,活泼好动,模型组毛色发黄,同时精神萎靡,金钗石斛多糖3个干预组以及贝那普利治疗组相较于模型组有一定好转;HE染色可见正常组心肌细胞排列整齐,而模型组心肌细胞明显肥大,同时肌纤维排列紊乱和断裂;金钗石斛多糖干预后,其心肌细胞排列明显好转,特别是高剂量组更为明显;与正常组相比,模型组血糖、CK、CK-MB、LDH、IL-1α、IL-1β、IL-6和TNF-α和胰岛素抵抗指数明显升高(P<0.05),胰岛素含量明显降低(P<0.05),金钗石斛多糖3个干预组和贝那普利治疗组相较于模型组血糖、CK、CK-MB、LDH、IL-1α、IL-1β、IL-6和TNF-α和胰岛素抵抗指数均明显降低(P<0.05),胰岛素含量则明显升高(P<0.05),而贝那普利治疗组变化趋势基本介于金钗石斛多糖低剂量组和中剂量组之间;与正常组相比,模型组RIP3表达明显升高(P<0.05),而金钗石斛多糖和贝那普利均可以降低其RIP3表达(P<0.05)。结论 :金钗石斛多糖可改善2型糖尿病心肌损伤大鼠心功能和炎症反应,此外对心肌细胞凋亡也有一定
Objective :To study and discuss the characteristics of heart function,inflammatory cytokines and RIP3 in heart tissue expression of type 2 diabetic rats treated with Dendrobium nobilepolysaccharide,provide some reference for the development of Dendrobium nobile polysaccharide and treating type 2 diabetes myocardial injury.Methods :60 SD rats were randomly divided into normal group,model group,Dendrobium nobile polysaccharide of low,middle and high dose intervention group,besides positive drug Benazepril treatment group,each group was 10 cases,excepted the normal group,the other 5 groups were given high fat diet for 6 months to built the rat model of type 2 diabetes.After successful builtding model,the normal group and model group were given 1 m L·kg^-1·d^-1 saline,Dendrobium nobile polysaccharide three intervention groups were given Dendrobium nobilepolysaccharides 100 mg·kg^-1·d^-1,200 mg·kg^-1·d^-1 and 400 mg·kg^-1·d^-1 treatment,Benazepril group were given Benazepril 1 mg·kg^-1·d^-1 by gavage treatment,the rats were killed after 8 weeks,HE staining detected the liver pathology,reagent for detected cardiac function and inflammatory factor,real-time quantitative RT-PCR and Western blot detected the expression level of RIP3 m RNA and protein.Results :The rats in the normal group was sleek,lively,model group hair color is yellow,and listlessness,compared with the model group,the Dendrobium nobile polysaccharide three intervention groups and Benazepril group had a certain improvement;HE staining could been see,the myocardium cells arranged was neat in the normal group,uniform size in the model group,the myocardial cells,muscle fiber hypertrophy,derangement and fracturein in the Dendrobium nobile polysaccharide group were significantly improved,while nuclear size was basically uniform,especially in the high dose group improved more significantly,in addition Benazepril group and Dendrobium nobile polysaccharide were the same dose group;compared with the normal group,model group CK,CK-MB,IL-1,LDH,IL-1�
