人参皂苷Rg3的抗乳腺癌转移作用研究 被引量:6

Inhibitory Effect of Ginsenoside Rg3 on Breast Cancer Metastasis

摘要 目的研究人参皂苷Rg3的抗乳腺癌转移的作用。方法 40只BALB/c小鼠采用尾静脉接种4T1乳腺癌细胞建立乳腺癌肺转移模型,随机分为4组,分别为模型对照组、人参皂苷Rg3低剂量组、中剂量组和高剂量组。各给药组每天灌胃给予相应剂量的人参皂苷Rg3,模型对照组灌胃给予生理盐水,每日1次,连续14天。以肺组织表面的肿瘤结节数、肺质量、血清MMP-2、MMP-9含量、肺组织VEGF含量为检测指标。结果与模型组比较,人参皂苷Rg3中剂量和高剂量组能够明显减少小鼠肺部的肿瘤结节数和降低肺组织质量(P<0.05或P<0.01);显著降低血清MMP-2和MMP-9的含量(P<0.05或P<0.01);显著减少肺组织中VEGF的含量(P<0.05或P<0.01)。结论人参皂苷Rg3可抑制乳腺癌肺转移,其作用机制可能与抑制MMP-2、MMP-9和VEGF的表达有关。 Objective To investigate the effect of ginsenoside Rg3 on breast cancer metastasis. Methods 40 female mice were randomly divided into 4 groups, including model group, ginsenoside Rg3 low dose, middle dose and high dose group. All of the animals were injected with 4T1 breast cancer cells via tail vein to establish a 4T1 routine model of breast cancer metastasis. After tumor inoculation, the mice were orally treated with ginsenoside Rg3 or vehicle for 14 days. The tumor nodules on the lungs, lung weight, the expression of MMP- 2 and MMP- 9 in serum and expression of VEGF in lung were measured. Results In ginsenoside Rg3 middle and high dose groups, the numbers of tumor nodules and lung weight were significantly reduced compared with model group (P〈0.05 or P〈0.01) ; the expression of MMP- 2 and MMP- 9 in serum and expression of VEGF in lung was significantly decreased compared with model group (P〈0.05 orP〈0.01). Conclusion Ginsenoside Rg3 has potent effect of inhibition of breast cancer metastasis, and the mechanisms may be involved with the inhibition of the expression of MMP- 2, MMP- 9 and VEGF.
