摘要
根据GenBank收录的马立克病病毒(MDV)基因组序列的Meq基因设计1对特异性引物,用PCR方法从河南发病鸡群中检测出了3株MDV,将目的片段克隆,并进行测序,获得Meq基因全长序列,将3株MDV的Meq基因与GenBank上收录的不同类型毒株进行比较分析。3株MDV的Meq基因段长度均为1 020bp,核苷酸序列分析表明,河南株Meq基因与其他MDV核苷酸同源性为98.6%,氨基酸同源性为96.5%;Meq基因的氨基酸序列共有9个位点的氨基酸存在突变;遗传进化分析表明这些毒株有3个分支,其中Henan 1~3株、ZC2014株(KP144356.1)、YLO40920株(DQ174459.1)、HNXZ106株(HF546099.1)和vv+MDV毒株648A(AY362725.1)位于同一个分支,vMDV GA(AF147806.1)、vvMDV RBIB(AY243332.1)位于同一个分支,mMDV CVI988(AF493555.1)和中国疫苗株814(AF493551)位于同一个分支。
To investigate genetic variation of Marek;s disease virus(MDV) in Henan province,one pair of primers for amplifying Meq gene of MDV were designed according to nucleotide sequence in GenBank,and three isolates of MDV were detected from infected chickens by PCR.Targeted fragments were cloned and sequenced,then compared with different types of reference MDV strains published in GenBank.The results showed that Meq gene from the three MDV isolates consisted of 1 020 bp.Compared with reference strains published in GenBank, the sequences of Meq gene in different isolates were relatively conserved and the homologies of nucleotide and amino acid sequences of the isolates were more than 98.6% and more than 96.5%.The isolates displayed amino acid site mutations at 9 positions. Phylogenetic analysis showed that the isolates and strains could be separated into three groups.The Henanl-3 strains,ZC2014 (KP144356.1), YLO40920 (DQ174459.1), HNXZ106 (HF546099.1) ,and vv+ MDV648A (AY362725.1) were clustered into one group,vMDV GA(AF147806.1),vvMDV RBIB (AY243332.1) clustered into the second group,and mMDV CVI988(AF493555.1) and Chinese vaccine 814 strains (AF493551) were clustered into the third group.
