酸王苹果被迫休眠期的活性氧代谢特征

摘要 以8年生生长势相对一致的晚萌发加工苹果品种酸王(Malus domestic cv.Avrolles)为试材,以正常萌发加工苹果品种瑞林(M.domestic cv.Judeline)为对照,测定自然休眠解除后被迫休眠期芽内活性氧产生速率、抗氧化酶活性及抗氧化物质含量的变化,探讨酸王在被迫休眠期活性氧代谢特征,为揭示酸王晚萌发的内在原因提供理论依据。结果表明,酸王和瑞林芽内O_(2·)^-的产生速率、H_2O_2含量变化、SOD、POD、CAT酶活性及抗氧化物质的变化趋势相同,但变化幅度有明显差异。在萌芽前,酸王和瑞林芽内的O_(2·)^-产生速率分别比自然休眠解除时提高了3.4倍和2.35倍,但酸王比瑞林低10.8%;酸王与瑞林芽内H_2O_2含量最高值分别为1.45和1.89OD_(410)/g FW,酸王低于对照瑞林;酸王芽内SOD、POD和CAT三种酶活性的最高点分别为瑞林的83.6%、67.7%和89.4%,抗氧化物质AsA含量酸王与瑞林在整个被迫休眠期分别增加了2.68和2.83μmol/g FW。这表明酸王活性氧代谢程度明显低于同时期的瑞林,这可能是酸王晚萌发的主要原因之一。 In order to provide theoretical basis for revealing the internal mechanism of late bud sprout of Malus domestic cv.Avrolles,the very late bud sprout cultivar of M. domestic cv.Avrolles and the control cultivar of M. domestic cv.Judeline were used as experimental materials to investigate the generation rate of reactive oxygen,the changes in the activity of antioxidant enzyme and the cotents of antioxidants during their imposed dormancy after natural dormancy was released.The two cultivars were eight years old,and had the same growth potential.The results showed that: the generation rate of superoxide anion free radical( O_(2·)^-),the changes in H_2O_2 content,the activity of SOD,POD and CAT and the contents of antioxidants in the two cultivars had the same trend,but the amplitude of variation was different in the two cultivars.The O_(2·)^- generation rate before bud sprout increased by 3.4 times and 2.35 times during the imposed dormancy of Avrolles and Judeline compared with that when the natural dormancy was released,respectively. However,the peak of O_(2·)^- generation rate in Avrolles was 10.8% lower than that in Judeline.The highest contents of H_2O_2 were 1.45 and 1.89 OD_(410)/g FW in Avrolles and Judeline. The SOD,POD and CAT activities in Avrolles were 83. 6%,67. 7% and 89. 4% of those in Juedline,respectively. The As A contents increased by 2. 68 μmol/g FW and 2. 83 μmol/g FW in Avrolles and Judelin. All these results indicated that the degree of active oxygen metabolism in Avrolles was lower than that in Judeline in the same period,this maybe one of the main reasons for late bud sprout of Avrolles.
