遥测技术对自发性高血压大鼠呼吸、体温和自主活动的监测及分析 被引量:1

Monitoring of Respiration, Body Temperature and Autonomic Activity in SHR by Implantable Telemetry

摘要 [目的]应用植入式遥测技术,观察清醒无束缚状态下自发性高血压大鼠(spontaneously hypertensive rat,SHR)呼吸、体温和自主活动的昼夜波动及不同月龄、性别间的差异。[方法]取3月龄SPF级SHR共16只,雌雄各半,手术植入C50-PXT遥测植入子,术后7 d开始观测。在3、5、7月龄时分别对清醒无束缚SHR呼吸、体温和自主活动进行24h连续监测。[结果]3月龄SHR夜间平均呼吸频率、体温和自主活动频率均高于白昼,差异有统计学意义(P<0.05)。夜间和白昼平均呼吸频率分别为(85.16±5.84)次/min和(78.66±5.67)次/min,8:00 PM和1:00 AM左右出现峰值,分别为(94.20±3.21)次/min和(93.06±4.11)次/min;2:00 PM^3:00 PM出现低谷,谷值为(71.21±2.37)次/min。夜间和白昼平均体温分别为(37.69±0.44)℃和(37.18±0.39)℃,在9:00 PM^10:00 PM和1:00 AM^2:00 AM左右出现峰值,分别为(38.09±0.31)℃和(38.11±0.33)℃;在2:30 PM^3:30 PM出现低谷,谷值为(36.72±0.21)℃。白天SHR自主活动频率差异大,在6:30 AM和8:00 AM左右出现两个峰值,分别为(47.43±5.42)次/min和(50.74±4.79)次/min。随着月龄的增加,SHR的呼吸频率逐渐降低;体温和自主活动则存在性别差异:雄性SHR体温逐渐升高,雌性则相反;雌性SHR的自主活动频率随月龄增加,不同月龄间比较,差异有统计学意义(P<0.05),而不同月龄雄性SHR的自主活动则无统计学差异(P>0.05)。[结论]3月龄SHR的呼吸、体温和自主活动呈昼夜节律性变化,夜间平均显著高于白昼。SHR呼吸频率随月龄的增加而逐渐减低,性别间差异不显著;随月龄增加,雄性和雌性SHR的体温分别呈现逐渐升高和降低的趋势;雌性自主活动随月龄显著增多,而雄性变化不大。 [Objective] To observe dynamic changes, sex differences and the circadian fluctuation of respiratory rate, body temperature and autonomic activity rate in unrestrained spontaneously hypertensive rat(SHR) by implantable telemetry. [Methods] Sixteen SPF grade SHR, half male and half female,were implanted with C50-PXT implantation, and experiment was started after the recovery of 7 d. Respiratory rate, body temperature and autonomic activity rate were respectively monitored for 24 h continuously using telemetry system when SHR were 3,5,7 months old. [Results] The mean frequency of respiration,body temperature and autonomic activity of SHR at night stage were significantly higher than day stage( P0.05). The average respiratory rate of night and day were(85.16±5.84) times/min and(78.66±5.67) times/min, respectively. There were two peaks of respiratory rate at 8:00 PM and 1:00 AM,(94.20 ±3.21) times/min and(93.06±4.11) times/min, respectively; and there was a low valley between 2:00 PM^3:00 PM,(71.21 ±2.37) times/min. The average body temperature at night and day were(37.69±0.44) degrees and(37.18±0.39) degrees, respectively. There were two peaks of body temperature between 9:00 PM^10:00 PM and 1:00 AM^2:00 AM(38.09±0.31) degrees and(38.11±0.33) degrees, respectively; and a valley period between 2:30 PM^3:30 PM,(36.72 ±0.21) degrees.During day stage, activity rate of SHR changed significantly, two peaks appeared at 6:30 AM and 8:00 AM, respectively(47.43 ±5.42) time/min and(50.74±4.79) time/min. SHR respiratory rate decreased as age increased. Male SHR body temperature rose as age increased, while female SHR was opposite. Male SHR autonomic activities rate had no obvious change as age increased; but female SHR activity rate significantly increased with the age( P 0.05).[Conclusion] Three-month-old SHR showed circadian rhythm changes of respiratory rate, body temperature and autonomic activity rate.
