高效液相色谱法测定金振口服液中6种成分含量 被引量:8

摘要 目的建立测定金振口服液中黄芩苷、芦荟大黄素、大黄酸、甘草酸、大黄素和大黄酚6种成分含量的高效液相色谱法。方法色谱柱为Phenomenex C_(18)柱(250 mm×4.6 mm,5μm),流动相为甲醇(A)-0.05%磷酸溶液(B),梯度洗脱,流速为1.0 m L/min,柱温为30℃,波长切换时间序列采样(黄芩苷0~15.0 min、280 nm,芦荟大黄素、大黄酸15.0~35.0 min、254 nm,甘草酸35.0~39.8 min、237 nm,大黄素、大黄酚39.8~50.0 min、254 nm)。结果黄芩苷、芦荟大黄素、大黄酸、甘草酸、大黄素和大黄酚进样量分别在0.043 2~2.162 5μg,0.012 8~0.638 0μg,0.018 5~0.926 0μg,0.035 8~1.792 5μg,0.049 8~2.488 0μg,0.052 9~2.643 5μg范围内与峰面积线性关系良好,平均加样回收率分别为98.84%,99.49%,99.89%,100.31%,99.55%,99.54%,RSD分别为1.10%,1.03%,1.49%,1.23%,0.98%,1.19%(n=9)。结论该方法简单快捷,准确可靠,可用于金振口服液的质量控制。 Objective To establish an HPLC method for simultaneous determination of baicalin, aloeemodin, rhein, glycirrizic acid, emodin and chrysophanol in Jinzhen Oral Solution. Methods The chromatographic column was Phenomenex C18 column(250 mm × 4. 6 mm, 5 μm), the mobile phase was methanol ( A ) -0. 05% phosphoric acid solution ( B ) with gradient elution, the flow rate was 1. 0 mL/min, the column temperature was 30 ℃, the detection wavelength was 280 nm for baicalin ( 0-15. 0 min ) , 254 nm for aloeemodin and rhein ( 15. 0-35. 0 min ) , 237 nm for glycirrizic acid ( 35. 0-39. 8 min ) , 254 nm for emodin and chrysophanol ( 39. 8-50. 0 min ) . Results The linear range of baicalin, aloeemodin, rhein, glycirrizic acid, emodin and chrysophanol were 0. 0432-2. 1625 μg, 0. 0128-0. 6380 μg, 0. 0185-0. 9260 μg, 0. 0358-1. 7925 μg, 0. 0498-2. 4880 μg, 0. 0529-2. 6435 μg, respectively. The average recovery rates were 98. 84%, 99. 49%, 99. 89%, 100. 31%, 99. 55%, 99. 54%, respectively, the RSDs were 1. 10%, 1. 03%, 1. 49%, 1. 23%, 0. 98%, 1. 19%( n=9 ) . Conclusion This method is simple and fast, which can be used for quality control of Jinzhen Oral Solution.
