浙江局部地区汉族人基质金属蛋白酶-3基因多态性与冠心病血瘀证的相关性研究 被引量:6

摘要 目的探讨基质金属蛋白酶-3(MMP-3)基因多态性与冠心病血瘀证的相关性。方法选择2013年9月至2015年9月浙江中医药大学附属第二医院、浙江省立同德医院心内科住院患者312例,通过冠状动脉造影检查将其中153例明确诊断为冠心病的患者,根据冠心病中医证型辨证分型为血瘀证组和非血瘀证组,其余159例设为对照组,采用聚合酶链反应-限制性片段长度多态性(PCR-RELP)检测受试者MMP-3-1171 5A/6A的基因型。结果 MMP-3一1612 5A/6A基因多态性与冠心病血瘀证发生相关,冠心病血瘀证组和对照组5A基因频率差异有统计学意义(P<0.05),冠心病血瘀证组和对照组5A/5A基因型频率差异有统计学意义(P<0.01)。结论 MMP-3-1612 5A/6A基因多态性与冠心病血瘀证发生相关,5A/5A基因型及5A等位基因可能增加浙江汉族局部地区人群患血瘀证的风险。 Objective To investigate the association of matrix metalloproteinase -3(MMP-3) gene polymorphism withblood stasis syndrome in coronary heart disease(CHD). Methods A total of 312 inpatients were enrolled and underwentcoronary angiography at the Second Affiliated Hospital of Zhejiang Medical University and Tongde Hospital of ZhejiangProvince from September 2013 to September 2015. CHD was determined in 153 cases and excluded in 159 cases(controlgroup) based on the results of coronary angiography. CHD patients were divided into blood stasis syndrome group andnon-stasis blood syndrome group according to TCM syndrome type of CHD. The genotypes of MMP-3-1171 5A/6A weredetermined by polymerase chain reaction and restriction fragment length polymorphism analysis. Results MMP-3-1612-5A/6A gene polymorphism was related to blood stasis syndrome of CHD. There were significant differences of 5Agene frequency and 5A/5A genotype frequency between blood stasis syndrome group and control group(P〈0.05, P〈0.01). Conclusion MMP-3-1612,-5A/6A gene polymorphism is associated with blood stasis syndrome of coronary heartdisease. 5A/5A genotype and 5A allele may increase the risk of blood stasis syndrome in the Chinese Han population in Zhejiang.
