摘要
目的探讨TOLL样受体3(TLR3)在大鼠心肌缺血/再灌注(I/R)损伤中的表达及作用机制。方法制备TLR3基因敲除大鼠及野生型大鼠,采用球囊结扎冠脉方法制作心肌缺血/再灌注动物模型,将60只大鼠随机分为假手术组(野生型)(n=15)、缺血/再灌注组(野生型)(n=15)、假手术组(TLR3-/-)(n=15)、缺血/再灌注组(TLR3-/-)(n=15)。检测各组大鼠的心肌组织MDA、GSH、CK-MB、LVESV、LVEDV、LVEF及TLR3、NF-κB表达。结果缺血/再灌注组(TLR3-/-)的MDA[(9.72±6.15)nmol/L]、CK-MB含量[(1075.76±3.27)U/L]低于缺血/再灌注组(野生型)[(14.85±5.2)nmol/L,(1842.16±4.28)U/L],GSH含量[(152.46±2.82)mg/L]高于缺血/再灌注组(野生型)[(128.72±1.82)mg/L],差异有统计学意义(P<0.05)。缺血/再灌注组(TLR3-/-)组的LVEDV[(602.0±2.7)μl]、LVESV[(516.0±2.9)μl]低于缺血/再灌注组(野生型)[(702.0±4.5)、(516.0±2.9)μl],LVEF[(42.0±2.8)%]高于缺血/再灌注组(野生型)[(36.0±4.2)%],差异有统计学意义(P<0.05)。缺血/再灌注组(野生型)TLR3(0.862±0.158)、NF-κB(0.786±1.784)蛋白表达均高于假手术组(野生型)(0.315±1.834,0.206±1.924),差异有统计学意义(P<0.05)。缺血/再灌注组(TLR3-/-)的TLR3蛋白(0.076±1.52)及NF-κB表达(0.625±2.824)低于缺血/再灌注组(野生型),差异有统计学意义(P<0.05)。结论 TOLL3受体基因缺失在心肌缺血损伤中具有心脏保护作用,其保护机制与减轻NF-κB,减轻心肌缺血/再灌注炎症反应有关。
Objective To investigate the expression of Toll-like receptors 3 and the correlation mechanism of ischemia/ reperfusion injury in ratsiethods TOLL-like receptor 3 deficient (FLR3-/-) and wild type (WT) rats were subjected to ischemia/ reperfusion induced by ligation of the left anterior descending coronary artery.Sixty rats were divided into 4 groups: sham-operated group (WT) (n=15),myocardial ischemia/reperfusion group (WT) (n=15),sham-operated group (TLR3-/-) (n=15) and myocardial isehemia/reperfasion group (TLR3-/-) (n=15).The MDA,GSH,CK-MB,LVEDV,LVESV,LVEF and the expression of TLR3,NF-κB in four groups were detecte.Results The level of MDA [(9.72±6.15)nmol/L],CK-MB [(1075.76±3.27)U/L] in myocardial ischemia/reperfusion group (TLR3-/-) were lower than the myocardial ischemia/reperfusion group (WT) [(14.85±5.2)nmol/L,(1842.16±4.28)U/L],the value of GSH in myocardial ischemia/reperfusion group (TLR3- /-) was higher than the myocardial ischemia/reperfusion group (WT),and the differences were statistically significant (P〈 0.05).The LVEDV [(602.0±2.7)μl],LVESV [(516.0_+2.9)μl] of the myoeardial ischemia/reperfusion group (TLR3-/-) were lower than the myocardial ischemia/reperfusion group (WT) [(702.0±4.5)μl,(516.0±2.9)μl],the LVEF [(42.0±2.8)%] of the myocardial isehemia/reperfusion group (TLR3-/-) was higher than the myocardial ischemia/reperfusion group (WT) [(36.0±4.2)% ],and the differences were statistically significant (P〈0.05).The expression of TLR3 (0.862±0.158),NF-κB (0.786±1.784) of the myocardial ischemia/reperfusion group (WT) were higher than the sham-operated group (WT) (0.315±l.834,0.206_+l.924),and the differences were statistically significant (P〈0.05).The expression of TLR3 (0.076± 1.52),NF-κB (0.625±2.824) of the myocardial ischemia/reperfusion group (TLR3-/-) were lower than the myocardial ischemia/reperfusio
