摘要
目的观察大蒜素对小鼠脾脏指数及其T淋巴细胞亚群分化的影响。方法将80只清洁级昆明小鼠随机化分组:生理盐水对照组、大蒜素低剂量组、大蒜素中剂量组和大蒜素高剂量组,每组20只,并分别按0.1ml/(10g bw)、100mg/(kg bw)、200mg/(kg bw)、400mg/(kg bw)的剂量连续灌胃14d,每天灌胃1次并称重。灌胃结束7d后处死小鼠,无菌分离脾脏称重,计算脾脏系数,制作单细胞悬液,采用流式细胞仪检测小鼠T淋巴细胞亚群中CD4^+T淋巴细胞、CD8^+T淋巴细胞和CD4^+/CD8^+的值。结果大蒜素中、高剂量组和生理盐水对照组比较,脾脏指数、脾脏淋巴细胞CD4^+T淋巴细胞比例和CD4^+/CD8^+值均升高,CD8^+T淋巴细胞比例降低,差异有统计学意义(F=5.52,P<0.05),而大蒜素低剂量组与生理盐水对照组比较,脾脏指数、脾脏淋巴细胞CD4^+T淋巴细胞、CD8^+T淋巴细胞和CD4^+/CD8^+值差异无统计学意义(F=1.87,P>0.05)。结论大蒜素能够使得小鼠脾脏指数、脾脏淋巴细胞CD4^+T淋巴细胞比例和CD4^+/CD8^+值均升高,CD8^+T淋巴细胞比例降低,对正常小鼠脾脏免疫功能具有正向调节作用。
Objective To observe the effects of allicin on spleen T lymphocyte subset in mice.MethodsEighty healthy adult Kunming mice were randomly and evenly divided into 4 groups,including normal saline control group,allicin low-dose group,mid-dose group and high-dose group,which were treated with normal saline and 0.1ml/（10g bw）of allicin at 100mg/（kg bw）,200mg/（kg bw）and 400mg/（kg bw）by gavage once a day for 14 days.They were sacrificed 7 days after the final gavage and the spleens were collected to obtain the spleen index and to make single cell suspension.FCM was used to determine the percentage of CD4^＋ and CD8^＋T lymphocyte and to calculate the CD4^＋/CD8^＋ ratio.Results The amount of CD4^＋T cells,CD4^＋ /CD8^＋ ratio and the spleen index were higher in the mid-dose and high-dose allicin mice group compared with normal saline control group,while the amount of CD8^＋T cells were lower,the differences were significant（F=5.52,P〈0.05）.However,the differences were not statistically significant when compared the lowdose group with the control group（F=1.87,P〉0.05）.Conclusions Mid-dose and high-dose allicin can upregulate CD4^＋T cells and down-regulate CD8^＋T cells and increase the CD4^＋ /CD8^＋ ratio in the spleen of mice.
