摘要
通过对西安市某污水处理厂活性泥的分离得到偶氮染料活性黑5的脱色菌株EWA2,经过生化试验和16S r DNA同源性比对分析,鉴定为奇异变形杆菌(Proteus mirabilis)。该菌在LB液体培养基中48 h内对300 mg/L以下的活性黑5脱色率高达94.7%~99.9%。在活性黑5模拟废水中,菌株EWA2可利用羧甲基纤维素钠和葡萄糖作为碳源显著提高对活性黑5的脱色率,在8%Na Cl条件下48 h内对活性黑5的脱色率达83.6%。该菌主要通过生物降解途径实现对活性黑5的脱色,降解产物的吸收峰位于近紫外光区。
A bacterial strain, EWA2, with remarkable ability to decolorize azo dye Reactive Black 5, was isolated from acti-vated sludge collected from a wastewater treatment plant in Xi'an. By means of biochemical test and homology comparison of16 S r DNA sequence, strain EWA2 was identified as Proteus mirabilis. In LB liquid medium, decolorization rate of Reactive Black 5(≤300 mg/L) by strain EWA2 could reached 94.7%~99.9% within 48 hours. In simulated wastewater containing Reactive Black 5, strain EWA2 could utilize sodium carboxymethyl cellulose and glucose as carbon source to significantly in-crease the decolorization rate. Under the condition in 8% Na Cl, the decolorization rate was 83.6% within 48 hours. It indicated that biodegradation was the main way of decolorizing Reactive Black 5 by strain EWA2, and the adsorption peak of degrada-tion products appeared in near-ultraviolet region.
