摘要
利用镰刀菌(Fusarium sp.)HJ01降解中性艳蓝GL,经扩增翻译延伸因子TEF1对HJ01菌株做了种的鉴定,并考察了温度、pH值对该菌株降解酶漆酶的活性,以及对中性艳蓝GL脱色率及COD去除率的影响.结果表明。该菌株屑层出镰刀菌(Fusarium proliferatum).HJ01菌株所产降解酶漆酶在pH为2.0-8.0内相对活性在12%-96%之间。在pH=4.0时,相对活性最大;在25-65℃内,随着温度的升高,漆酶相对活性增大;在65-85℃内,随着温度增大,漆酶相对活性降低.Hj01菌株在25气、50r·min^-1条件下,于液体培养基中培养96h后加入到25mL含100mg·L^-1中性艳蓝GL的溶液中,在35℃、50r·min^-1、pH=4.0条件下继续培养100h后,中性艳蓝GL的脱色率达100%,培养至120h后,COD去除率选87.8%.紫外光谱分析表明,中性艳蓝GL的发色基团蒽醌环被漆酶打破.
The study on neutral brilliant blue GL decoloration and degradation characteristics was carried out by the Fusarium sp. HJ01 strain. The HJ01 was identified as Fusarium proliferatum through TEF1 (translation elongation factor 1 alpha). The effects of temperature and pH value on neutral brilliant blue GL decoloratian rate and COD removal rate were investigated. The results showed that the relative activity of laccase was between 12% and 96% in pH range from 2.0 to 8.0, and reached its peak at pH = 4.0. The relative activity increased with temperature increasing in the range from 25 ℃ to 65 ℃ , hut it was opposite when from 65 ℃ to 85 ℃. The H J01 mycelia incubated in the liquid culture for 96h at 25 ℃ and 50 r·min^-1 were added to the neutral brilliant blue GL solution (100mg·L^-1 , 25mL), and were cultured continuously for another 100h at 35 ℃ ,50 r·min^-1 and pH = 4.0, the decoloration rate of neutral brilliant blue GL was up to 100% , after 120h, COD removal rate was up to 87.8%. The ultraviolet - visible spectrum showed that the chromophore anthraquinonc ring of neutral brilliant blue GL was broken down by laccase.
