响应面法优化黄精黄酮提取工艺及其抗氧化活性研究 被引量:15

摘要 以九华山所产的多花黄精为材料,对其黄酮提取过程中的溶剂种类及其浓度、料液比、提取时间、提取温度以及提取次数进行单因素实验;在单因素实验的基础上利用Box-Behnken响应面实验优化黄精黄酮提取工艺;并对黄精黄酮的DPPH自由基和羟自由基的清除能力进行研究.结果表明,较为适宜的黄精黄酮提取工艺条件为乙醇体积百分比浓度为81%、提取温度为69℃、料液比为1∶18、提取时间为2.5h、提取次数为2次;此时黄精黄酮提取率的最大理论估计值为0.63%,在此条件下进行验证实验,测得黄酮实际的提取率为0.60%,与理论值比较接近.黄精黄酮具有很强的抗氧化活性,当其浓度为120μg/mL时,其DPPH自由基和羟自由基的清除率均与Vc相当. Using Polygonati Rhizoma as material, the extraction conditions of flavonoids including the solvent types and their concentration, solid-liquid ratio, extraction time, extraction temperature and ex- traction times were studied by single factor experiment.Meanwhile, the optimum process for flavonoids extraction .were researched by RSA method.In addition,the antioxidant activities of flavonoids were eval- uated by DPPH and · OH free radical scavenging assays.The results showed that the optimal extraction conditions were ethanol concentration of 81%, extraction temperature of 69 ℃, solid-liquid ratio of 1 : 18, extraction time of 2.5 h, and the repeated extraction number of 2.Under these conditions, the predic- ted and experimental extraction yield of flavonoids was 0.63 % and 0.60 %, respectively.The flavonoids in Polygonati Rhizoma showed strong antioxidant activity,and the scavenging rate of DPPH and · OH were equivalent to that of Vc when flavonoids concentration was 120μg/mL.
