摘要
运用高速逆流色谱技术从铁皮石斛中分离制备出芥子酸,为石斛属内首次分离得到。分离采用的溶剂体系为石油醚-乙酸乙酯-甲醇-水(4∶6∶3∶7,体积比),上相做固定相,下相做流动相,流速2 m L/min,转速800 r/min,检测波长280 nm。单次进样石斛提取物200 mg,分离出纯度为93.11%的芥子酸20.34 mg,固定相保留率46.67%。体外抗氧化结果显示,芥子酸有较强的DPPH自由基清除能力,IC50为0.046 mg/m L。
Sinapic acid was separated and purified from Derdrobium officinale by high-speed countercurrent chromatography, which was isolated for the first time from Dendrobium. Ether-ethyl acetate-methanol-water (4 ∶ 6 ∶ 3 ∶ 7, volume ratio) was used as the two-phase solvent system, in which the upper and lower phase were used as the stationary and mobile phase, respectively. The flow rate was set at 2 mL/min, and the revolution speed was 800 r/min. The detection wavelength was 280 nm and the retention of stationary phase was 46.67 %. Finally, 20.34 mg of sinapic acid with a purity of 93.11 % was obtained in a one-step separation from 200 mg of crude extracts. The results showed that sinapic acid had a strong DPPH radical scavenging ability in vitro and the IC50 was 0.046 mg/mL.
