摘要
目的:研究大蒜素对子宫内膜癌Ishikawa细胞增殖和凋亡的影响,并探讨其可能的作用机制。方法:以不同浓度大蒜素(12.5、25、50μg/mL)和顺铂(40μg/mL)分别干预对数生长期Ishikawa细胞,采用MTT法检测Ishikawa细胞增值抑制;通过流式细胞仪检测细胞周期和细胞凋亡状况,采用Western blotting法检测Ishikawa细胞中caspase-3蛋白表达,RT-PCR法检测Ishikawa细胞中Bax mRNA、bcl-2 mRNA表达并计算Bax/bcl-2表达比值。结果:大蒜素能够显著提高子宫内膜癌Ishikawa细胞增殖抑制率,阻滞Ishikawa细胞周期于G2/M期,提高细胞凋亡率,上调caspase-3蛋白和Bax mRNA表达,下调bcl-2 mRNA表达,提高Bax/bcl-2表达比值,且上述作用均具有一定的剂量依赖性。结论:大蒜素对子宫内膜癌Ishikawa细胞增殖具有抑制作用以及促其凋亡的作用,机制可能与大蒜素能够改变Ishikawa细胞周期以及调节凋亡相关基因、蛋白表达有关。
Objective: To investigate the effects of allimin on the proliferation and apoptosis of endometrial carcinoma Ishikawa cell, and to discuss its possible mechanism. Methods: The endometrial carcinoma Ishikawa cells in logarithmic phase were intervened with allimin in different concentrations(12.5, 25 and 50 μg/m L) and cis-platinum complexes(40 μg/m L). Cellular growth inhibition rate was detected by MTT method; cell cycle and apoptosis rate were analyzed by flow cytometry, the expression of caspase-3 protein was detected by Western blotting method,and the expressions of Bax mRNA and bcl-2 mRNA in the Ishikawa cells were detected by RT-PCR method, and the ratio of Bax/bcl-2 was calculated. Results: Allimin could remarkably raise the inhibition rate of Ishikawa cells proliferation, arrestthe cell cycle at G2/M phase, increase cellular apoptosis rate, up-regulate the expressions of caspase-3protein and Bax mRNA,down-regulate the expressions of bcl-2 mRNA and improve the ratio of Bax/bcl-2, and the effects above showed dose-dependent to some extent. Conclusion:Allimin could inhibit the proliferation of endometrial carcinoma Ishikawa cells and promote the apoptosis, which perhaps might be related to its effects of changing the Ishikawa cell cycle and regulating the apoptosis-related gene and protein expressions.
