摘要
目的:观察灯盏细辛注射液对动脉硬化家兔脂代谢、内皮功能及基质金属蛋白酶9/基质金属蛋白酶抑制因子-1(MMP-9/TIMP-1)影响。方法 :60只雄性SD家兔随机分为对照组、模型组、他汀组、灯盏低剂量和高剂量组,采用高脂喂养联合耳缘静脉注射小牛血清白蛋白方法建立动脉硬化模型。实验第10周、18周时,酶法检测内皮功能和血脂水平,免疫组织化学法检测MMP-9、TIMP-1的表达,组织形态学方法分析斑块情况。结果:第10周和18周,与对照组比较,模型组和各给药组内皮素1(ET-1)、总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、MMP-9水平和斑块面积比明显升高,而高密度脂蛋白胆固醇(HDL-C)、一氧化氮(NO)、TIMP-1水平明显降低(均P<0.05),可见明显斑块形成。第18周,与模型组比较,各给药组ET-1、TC、TG、LDL-C、MMP-9水平和斑块面积比明显下降,HDL-C、NO、TIMP-1水平明显升高(均P<0.05),斑块明显减轻。并且灯盏细辛注射液剂量越高,改变越明显。结论:灯盏细辛注射液可以减轻动脉粥样硬化家兔斑块,可能与其调控脂质代谢、改善内皮功能、调节MMP-9/TIMP-1动态变化有关。
Objective：To investigate the effects of Dengzhanxixin injection on lipid metabolism,vascular endothelial function and MMP-9/TIMP-1in rabbit models with experimental atherosclerosis.Method：Sixty healthy adult male rabbits were randomly divided into the control group,the model group,the atorvastatin group,the groups of low and high dose of Dengzhanxixin injection.The atherosclerosis rabbit models were induced by feeding high-fat diet plus injecting bovine serum albumin.At 10 and 18weeks,enzymology method was used to detect the levels of serum lipid and vascular endothelial function.The expressions of MMP-9and TIMP-1in aorta were observed by immunohistochemistry technique.The plaque area of aorta was performed with histomorphometry analysis.Result：At 10 and 18weeks,compared with the control group,the levels of ET-1,TC,TG、LDL-C,MMP-9significantly increased,but HDL-C,NO and TIMP-1markedly decreased with up-regulated arota atherosclerotic plaques and patch area ratio（all P〈0.05）.However,at 18 weeks,all above effects were significantly reversed by treatment with Dengzhanxixin injection（all P〈0.05）.Conclusion：Dengzhanxixin injection could decrease atherosclerosis by downregulating ET-1,TC,TG,LDL-C,MMP-9and upregulate HDL-C,NO,TIMP-1.
