摘要
针对市售枸杞提取物中多糖含量难以确定的问题,建立了先用淀粉葡萄糖苷酶脱除麦芽糊精,再用高效凝胶色谱分离定量的检测方法,经分析验证淀粉葡萄糖苷酶降解麦芽糊精的同时对枸杞多糖含量并无显著显影响,高效凝胶色谱可以实现不同分子量范围多糖的精确定量。检测结果表明,枸杞提取物中多糖含量为12%,相对分子质量范围为10 000~60 000 U,低聚糖含量较高为51.3%。
It was to develop a method for determination of polysaccharide in sold extract of Lycium barbarum,to degrade maltodextrin using amyloglucosidase,to separate and detect polysaccharide by high performance gel permeation chromatography(HPGPC).It was testified that polysaccharide couldn't be significantly influenced at the same time.And the molecular mass range and content of polysaccharide in sold extract of Lycium barbarumL.were determined.The molecular weight distribution of polysaccharide in extract of Lycium barbarumL.was 10 000-60 000 U,content was 12%,and the content of oligosaccharide was 51.3%.
