摘要
采用超临界流体技术制备白术内酯脂质体,并对其进行表征。将脂质体膜材与白术内酯溶解在超临界CO_2/乙醇中,经膨胀后快速喷射在缓冲溶液中,脂质载体自组装形成载药脂质体。考察了超临界参数对脂质体制备的影响,测定了包封率与平均粒径,表征了脂质体的微观形貌、粒度分布、物相特征。结果表明,在压力为30 MPa,温度为338 K,x(CH_3CH_2OH)=15%条件下,白术内酯以无定形态包封于磷脂载体中,形成了白术内酯脂质体。所制备脂质体白术内酯包封率达到80%以上,平均粒径为100 nm左右,粒度呈正态分布,其质量标准符合食药监局关于纳米药物/食品粒子的要求,为进一步将其制成纳米食品提供了功能原料。
The liposome entrapping atractylenolide was formed by supercritical carbon dioxide process and characterized.After liposomal materials and atractylenolide were dissolved in the mixture of supercritical carbon dioxide(SC-CO_2)/ethanol,the solution was expanded and sprayed into aqueous medium.The physicochemical properties including entrapment efficiency and average size were measured.The micromorphology,particle size distribution,phase characteristics of liposome were characterized.When the SC-CO_2/ethanol solution(x(CH_3CH_2OH)=15%) was expanded into water at pressure of 30 MPa,temperature of 338 K,the entrapment efficiency and average particle size of liposome were found to be above 80% and about 100 nm,respectively,which achieved the standards of Chinese pharmacopoeia.These results indicated that the liposome entrapping atractylenolide was suitable for development of nono-functional food.
