扶正透毒祛毒复方含药血清干预T-ALL-CR患者骨髓CD34^+细胞源DC诱导的研究 被引量:2

Effect of Serum in Compound Fuzheng Toudu Qudu on CD34^+ Cell Dendritic Cells in Patients with T-ALL-CR

摘要 目的:探讨加味青蒿鳖甲汤对干预诱导的T-ALL-CR患者骨髓CD34^+细胞源树突细胞的影响。方法:分别用高、中、低剂量加味青蒿鳖甲汤煎剂灌胃健康新西兰大白兔制备高、中、低浓度的含药血清;用Ficoll密度梯度离心法分离T-ALL-CR患者骨髓获得骨髓单个核细胞(Bone Marrow Nucleated Cell,BMNC),用CD34^+免疫磁珠阳性选择法提取BMNC中CD34^+细胞;将CD34^+细胞加入含细胞因子的培养体系中培养诱导DC,并分组加入高、中、低剂量含药血清干预诱导,诱导过程中采用倒置显微镜观察细胞形态并拍照记录,9天后收集细胞用流式细胞仪(Flow Cytometry,FCM)检测培养细胞CD80、CD86、CD83、CD1a、HLA-DR的表达以鉴定DC的成熟。结果:DC形态随着培养时间的延长,观察发现CD34^+细胞逐渐聚集,出现不规则突起,形成集落,可见较为典型的DC形态;诱导第9天后CD80、CD86、CD83、CD1a、HLA-DR所有表面抗原在各组中均有较高表达,不同剂量含药血清组的表达大多数优于正常兔血清组及细胞因子组(P<0.05),不同剂量含药兔血清组间的表达无明显差异(P>0.05)。结论:不同剂量加味青蒿鳖甲汤含药血清能促进T-ALL-CR患者CD34^+细胞源DC的成熟,能较高表达DC表面抗原CD83、CD86、CD80、CD1a、HLA-DR。 Objective:To investigate the effect of serum in compound Fuzheng Toudu Qudu modified with preparation Qinghao Biejia Decoction on CD34+Cell Dendritic cells in patients with T-ALL-CR. Methods:Healthy New Zealand white rabbits were gavaged with high,medium and low doses of modified preparation Qinghao Biejia Decoction respectively to prepare high,medium and low concentrations of drug-containing serum. Bone Marrow Nucleated Cell( BMNC) was obtained from the bone marrow of patients with T-ALL-CR by Ficoll density gradient centrifugation,and CD34+ cells were extracted from BMNC by CD34+ immunomagnetic beads. CD34+ cells were added to the culture system containing cytokines to induce DCs,and then divided into groups with high,medium and low doses of drug-containing serum intervention,meanwhile,inverted microscope was used to observe the cell morphology and photographed recording. The cells were collected after 9 days and the Flow Cytometry( FCM) was used to detect the expression of CD80,CD86,CD83,CD1 a and HLA-DR in cultured cells to identify the maturation of DCs. Results: With the prolongation of culturing time,CD34+ cells gathered gradually,irregular protrusions appeared and colonies formed,which showed a typical DC morphology. The expressions of all the surface antigens of CD80,CD86,CD83,CD1 a and HLA-DR were higher in all groups on the 9 th day after induction,and most of the expressions in the different doses of the drug-containing serum group were better than those in the normal rabbit serum group and the cytokine group( P 〈0. 05). There was no significant difference between the two groups( P 〉 0. 05). Conclusion: The serum in different doses of modified preparation Qinghao Biejia Decoction can promote the maturation of DCs derived from CD34+ cells in T-ALL-CR patients,and can express DC surface antigen CD83,CD86,CD80,CD1 a and HLA-DR.
