摘要
以苦荞麦蛋白为原料,经过酶解制备出苦荞麦活性肽,并采用HPLC将提取的苦荞麦活性肽进行纯化分离,制备出3个不同的肽段(5.8、7.1、32 min 3个峰值的样品)。采用DPPH、ABTS、羟基自由基3种方法检测苦荞麦3个不同肽段的抗氧化效果。试验表明,分离出来的3个肽段都具有抗氧化能力。对DPPH自由基的清除作用中,3个活性肽浓度均为0.1 mg/m L时,清除率分别为48.42%、62.06%、56.87%;对OH自由基的清除作用中,3个活性肽浓度为0.1 mg/m L时,清除率分别为45.07%、63.29%、48.10%;在对ABTS^+自由基的清除作用中,3个活性肽浓度为0.1 mg/m L时,清除率分别为53.07%、69.32%、46.01%。结论:分离出来的3个肽段都具有较强的抗氧化能力,其中以7.1 min收集的样品2对DPPH自由基、羟基自由基、ABTS^+自由基的抗氧化作用最强,均达到60%以上。
The bioactive peptides was extracted from protein of tartary buckwheat by enzyme method.And using the HPLC to separation and purification tartary buckwheat bioactive peptide, were prepared by three different peptides(5.8, 7.1, 32 min three peak samples). The DPPH, ABTS and hydroxyl free radical methods were used to detect the antioxidant effect of three different peptides from tartary buckwheat. The experiment showed that three different peptides in tartary buckwheat had the ability of antioxidation. In the scavenging action of DPPH free radical, the clearance rates were 48.42%, 62.06%, 56.87%, respectively, when the concentration of that three different peptides was 0.1 mg/m L. The clearance rate of OH free radical was 45.07%, 63.29%, 48.10%, respectively, when the concentration of that three different peptides was 0.1 mg/m L. In the scavenging effect on ABTS free radical, the concentration of that three different peptides was 0.1 mg/m L, and the removal rate was 53.07%, 69.32%, 46.01% respectively. Conclusion: The three peptides had strong antioxidant activity. Among the samples collected by 7.1 min, the antioxidant activity of DPPH free radical, hydroxyl radical and ABTS~+ radical was the strongest, which reached more than 60%.
