水稻抽穗期基因EHD8的遗传分析及精细定位 被引量:4

摘要 抽穗期是水稻重要的农艺性状之一,挖掘特异等位变异形式的抽穗期相关基因,对于改良水稻抽穗期至关重要。通过EMS诱变籼稻品种9311获得一个稳定遗传的早抽穗突变体ehd8。与野生型9311相比,ehd8在南宁自然长日照和南宁自然短日照条件下,分别提前抽穗6.7d和10.2d。此外,在南宁自然短日照条件下,ehd8的株高和每穗粒数比野生型9311显著降低。遗传分析表明,ehd8早抽穗性状受一对隐性核基因控制。EHD8基因被定位在8号染色体短臂上17kb范围内。基因注释信息表明,该定位区间内只有1个预测的候选基因LOC_Os080174500与已知的抽穗期基因EF8等位。序列比对分析发现,与野生型9311相比,ehd8突变体中LOC_Os080174500编码区的第11位碱基发生了G→T置换,造成其所编码蛋白的第四位氨基酸发生了精氨酸→蛋氨酸的置换。EHD8新等位基因的发掘对于丰富水稻特异种质资源以及促进其在水稻育种中的应用具有重要意义。 Heading date is one of the important agronomic traits in rice. Allele mining of heading related genes is important for rice improvement. An early heading mutant 8 (EHD8) derived from an EMS-induced population of indica variety 9311 was isola-ted. Under natural long-day and short-day in Nanning, the heading date of EHD8 was 6. 7 d and 10. 2 d shorter than that of 9311,respectively. Moreover, EHD8 displayed decreased plant height and less grains per main panicle than 9311 under natural short-day in Nanning. Genetic analysis demonstrated that the early heading phenotype was controlled by a single recessive nuclear gene, which was fine-mapped to a 17 kb region of chromosome 8. The gene annotation indicated that there was only one putative gene LOC_Os0801 74500 in this region, which was allelic to EF8. Sequence analysis indicated that a single base substitution (G→T) at position ii bp from the start codon was detected in LOC_Os080174500, which substitute arginine for methionine. The i-dentification of EHD8 provides useful experimental materials and facilitates the application of EHD8 in rice breeding.
