摘要
采集10份健康奶牛阴道临床样品,利用MRS琼脂培养基进行细菌分离培养与纯化,通过形态特征观察、生化试验、16S r RNA基因序列对分离菌株进行鉴定,并通过抑菌试验检测分离菌株对大肠杆菌、金黄色葡萄球菌、沙门菌等致病菌的抑菌能力。结果显示,分离筛选到3株乳酸菌,其菌落特征、形态特征、生化特性均符合乳酸杆菌的特性,均具有较好的生长性能和较强的产酸能力,与Gen Bank中乳酸杆菌的同源性均在99%以上,对病原菌的抑菌环直径均在11.41 mm以上。研究表明分离鉴定的3株乳酸杆菌是有潜质的有益菌株,可作为奶牛子宫内膜炎防治的候选益生菌。
This study collected 10 healthy cow vaginal clinical samples,and isolated and identified bacteria based on morphological observation,biochemical test and 16 S r RNA gene sequences. Also,antibacterial activities against Escherichia coli,Staphylococcus aureus and Salmonella were determined. The results showed that three bacterial strains are characteristic of Lactobacillus in colony morphology and biochemical characteristics,and have a better growth performance and strong ability of acid production. The 16 S r RNA gene sequences of the three isolates have above 99% of homology with those of Lactobacillus strains published in Gen Bank. The inhibition diameters against E. coli,S. aureus and Salmonella were more than 11. 41 mm. This study indicated that the three isolates of Lactobacillus are potential beneficial bacteria,and can be used as the candidate probiotics to control endometritis in dairy cows.
