摘要
目的:探讨槐定碱对人胰腺癌细胞株capan-1增殖及NF-κB信号通路的影响。方法:采用不同浓度的槐定碱处理体外培养的胰腺癌capan-1细胞,采用MTT法检测细胞增殖的变化;Hoechst 33342染色法检测capan-1细胞凋亡;Western blot法检测细胞中NF-κBp65和IκB-α蛋白表达水平;ELISA法检测TNF-α、IL^(-1)β、IL-6表达水平。结果:与阴性对照组比较,不同浓度的槐定碱均可明显抑制人胰腺癌capan-1细胞增殖。在0.625,2.5,5 g·L^(-1)浓度下,槐定碱能够促进人胰腺癌capan-1细胞凋亡(P<0.05)。在实验条件下,NF-κBp65、TNF-α、IL^(-1)β、IL-6表达水平明显减低,IκB-α蛋白表达水平明显增加(P<0.05)。结论:槐定碱能抑制胰腺癌细胞株capan-1增殖,促进其凋亡,其机制可能与下调NF-κBp65、TNF-α、IL^(-1)β、IL-6表达,增加IκB-α表达,诱导细胞凋亡有关。
OBJECTIVE To explore the effects of sophoridine on proliferation and signaling transduction of NF-κB in human pancreatic cancer capan-1 cells.METHODS After capan-1 cells were incubated with various concentrations of sophoridine,cell proliferation was assessed by MTT assay.Cell apoptosis was detected by Hoechst 33342 staining.Expression of NF-κBp65 and IκB-αprotein was determined by Western blot assay.Expression levels of TNF-α,IL^-1βand IL-6 were determined by ELISA assay.RESULTS Compared with negative control group,sophoridine at different concentrations obviously inhibited proliferation of capan-1 cells.Apoptosis rates increased in capan-1 cells after being treated by sophoridine at concentrations of 0.625,2.5,5 g·L^-1(P〈0.05).Besides,sophoridine also decreased the expression of NF-κBp65,TNF-α,IL^-1βand IL-6 and increased the expression of IκB-α.CONCLUSIONSophoridine can inhibit cell proliferation,and prompt apoptosis in capan-1 cells,which may be associated with down-regulation of NF-κBp65,TNF-α,IL^-1 and IL-6,and upregulation of NF-κB levels.
