摘要
目的:观察红景天苷对糖尿病模型大鼠学习记忆能力及氧化应激的影响。方法:培养50只清洁级健康雄性Wistar大鼠,取其中30只分为正常组、糖尿病组及红景天苷组,每组10只。糖尿病组及红景天苷组予以腹腔注射链脲佐菌素(STZ)诱导制备糖尿病模型,正常组不予处理。红景天苷组大鼠予以50 mg·kg-1红景天苷灌胃治疗,连续治疗12周。观察并记录大鼠血糖、体质量、血清及肾脏组织超氧化物歧化酶(superoxide dismutase,SOD)及丙二醛(malonaldehyde,MDA)水平变化,同时采用Morris水迷宫实验测试大鼠学习记忆能力。结果:与糖尿病组比较,红景天苷组血糖水平较低[(16.43±2.34)mmol·L^(-1)],体质量水平[(241.86±33.12)g]较高,差异均有统计学意义(P<0.05);与糖尿病组比较,红景天苷组逃避潜伏期时间[(19.43±2.77)s]较短,达中心区域的比例[(26.37±3.76)%]较高,穿越目标区次数[(3.05±0.43)次]较多,差异均有统计学意义(P<0.05);与糖尿病组比较,红景天苷组血清及肾脏组织SOD水平[(132.45±18.92)U·m L^(-1)、(34.54±4.93)U·m L^(-1)]较高,MDA水平[(6.43±0.91)μmol·L^(-1)、(3.43±0.48)μmol·L^(-1)]较低,差异均有统计学意义(P<0.05)。结论:红景天苷能有效改善糖尿病模型大鼠学习记忆能力,升高血清及肾脏组织SOD水平,降低血清及肾脏组织MDA水平。
Objective : To investigate the effect of the learning and memoP/ability and oxidative stress by rhodioside in diabetic mod- el rats. Method:50 healthy male Wistar rats were cultured,30 of them were divided into three groups:normal group, diabetic group and salidroside group, each group had 10 rats. Diabetic group and salidroside group were injected with streptozotocin (STZ) to in- duce diabetic model, and the normal group was not treated. Salidroside group rats were treated with 50 mg - kg I salidroside for continuous treatment for 12 weeks. The changes of blood glucose, body weight, serum superoxide dismutase (SOD) and malondial- dehyde (MDA) in rats were observed and recorded,and the ability of learning and memory was tested by Morris water maze test. Results:Compared with the diabetic group,the blood glucose level was lower[ (16.43 ± 2.34)mmol·L(-1) and the body weight [ (241.86 ± 33.12) g] was higher in the salidroside group( P 〈 0.05 ) ; compared with the diabetic group, the escape latency was shorter[ ( 19.43 ± 2.77 ) s ], the proportion of the central area was higher[ (26.37 ± 3.76)% ], the number of crossing the target area was higher[ (3.05 ± 0.43 ) times l , and the difference was statistically significant (P 〈 0.05 ). Compared with the diabetic group,SOD level in serum and kidney tissue of salidroside group were higher [ ( 132.45 ± 18.92) mmol·L(-1), ( 34.54 ± 4.93 ) mmol·L(-1),and the levels of MDA were lower[ (6.43 ± 0.91 )mmol·L(-1), (3.43 ± 0.48 )mmol·L(-1) ,which had statistically sinificance(P 〈 O. 05 ). Conclusion : Salidroside can effectivelv improve the learning and memory ability of diabetic rats, increasethe level of SOD in serum and kidney tissue, and decrease the level of MDA in serum and kidney tissue.
