地龙组分对UUO大鼠BMP-7、Smad6、TGF-β_1表达的影响 被引量:11

摘要 目的:研究地龙组分对单侧输尿管结扎大鼠BMP-7、Smad6、TGF-β_1表达的影响,探讨地龙组分延缓肾纤维化的可能机制。方法:健康雄性wistar大鼠60只,随机分组:假手术组+模型组+地龙低剂量组+地龙高剂量组。采用单侧输尿管结扎法复制肾间质纤维化大鼠模型,取梗阻侧肾组织标本,免疫组织化学方法检测肾组织BMP-7、Smad6、TGF-β_1的表达。结果:地龙治疗组大鼠的肾间质纤维化有所改善;与模型组相比,地龙治疗组大鼠肾组织BMP-7、Smad6表达增多,TGF-β_1表达下降(P<0.05)。结论:地龙组分通过上调BMP-7、Smad6表达,下调TGF-β_1的表达,来延缓肾纤维化的进程。 Objective: To investigate the effete of Di Long compostion on the expression of bone motphogenetic protein-7( BMP-7) and Transforming growth factor-β_1( TGF-β_1) and Smad6 in renal tissue of rats by unilateral ureteral obstruetion( UUO),and to illustrate the mechanism of Di Long compostion on delaying renal interstitial fibrosis. Methods: In the study,60 male wistar rats were randomly divided into groups in cluding sham-operated group,model group,low-dose group of Di Long Capsule,high-dose group of Di Long Capsule. the rats by unilateral ureteral obstruetion( UUO) was adopted. The expression of BMP-7、Smad6 and TGF-β_1 in renal tissue were determined by immuno histoehemistry with two steps. Results: renal interstitial fibrosis of the rats treated by Di Long compostion was improved,the expression of TGF-β_1decreased and the expression of BMP-7 and Smad6 increased significantly compared with model group. Conclusion: Di Long compostion can decrease the expression of TGF-β_1and increase the expression of BMP-7 and Smad6 in renal tissue of UUO rats,it is possible mechanism of improving renal interstitial fibrosis.
