上调ob mRNA对慢性肾衰竭模型大鼠营养不良的影响及其机制 被引量:2

摘要 目的研究上调瘦素(ob)mRNA对慢性肾衰竭模型大鼠的干预效果及其作用机制。方法选择40只SPF级SD健康大鼠,其中正常组不作任何处理,建立慢性肾衰竭模型,建模成功后分为模型组、对照组、上调组各10只。进行细胞转染建立稳定转染的ob mRNA上调组。对4组大鼠病理组织苏木精-伊红(HE)染色、NO、NOS、Alb、Hb水平、SOD、MDA水平、ob mRNA表达进行检测,并作组间比较。结果上调组、对照组NO、NOS水平低于模型组,Alb、Hb水平高于模型组,差异有统计学意义(P<0.05);上调组NO、NOS水平低于对照组,Alb、Hb水平高于对照组,差异有统计学意义(P<0.05);上调组、对照组SOD、MDA、Scr、BUN水平低于模型组,差异有统计学意义(P<0.05);上调组SOD、MDA、Scr、BUN水平低于对照组,差异有统计学意义(P<0.05);上调组、对照组ob m RNA水平高于模型组,差异有统计学意义(P<0.05);上调组ob mRNA水平高于对照组。结论上调ob mRNA对慢性肾衰竭大鼠干预效果显著,能够有效降低大鼠的炎性介质水平,减少氧自由基的损伤,从而能够在一定程度上增强对肾脏的保护作用。 Objective To study the intervention effect and mechanism of upregulation of ob mRNA in rats with chronic renal failure.Methods Forty SPF grade SD healthy rats were selected,among which the normal group received no treatment,and the model of chronic renal failure was established.After successful modeling,the model group,the control group and the upregulated group were divided into 10 rats respectively.Cell transfection was carried out to establish a stable transfected ob mRNA upregulation group.The expressions of HE staining,NO,NOS,iNOS,eNOS,SOD,MDA and ob mRNA in four groups of rats were detected and compared.Results The levels of NO and NOS in the upregulation group and the control group were lower than those in the model group,and the levels of Alb and Hb were higher than those in the model group,showing a statistical difference(P<0.05).The levels of NO and NOS in the upregulated group were lower than those in the control group,and the levels of Alb and Hb were higher than those in the control group,showing a statistical difference(P<0.05).The levels of SOD,MDA,Scr and BUN in the upregulation group and the control group were lower than those in the model group,showing statistical differences(P<0.05).The levels of SOD,MDA,Scr and BUN in the upregulation group were lower than those in the control group(P<0.05).The mRNA levels of ob in the upregulation group and control group were higher than that in the model group,showing a statistical difference(P<0.05).The mRNA level of ob in the upregulation group was higher than that in the control group.Conclusion Upregulation of ob mRNA has a significant effect on the intervention of chronic renal failure rats,which can effectively reduce the level of inflammatory mediators in rats and reduce the damage of oxygen free radicals,thus enhancing the protective effect on the kidney to a certain extent.
