摘要
目的:建立具有中医证候特点的肝郁血瘀大鼠模型,探究醋香附治疗肝郁血瘀的有效物质基础,筛选其疏肝散瘀有效部位。方法:采用外界随机刺激造模法及盐酸肾上腺素所致的肝郁血瘀造模法,从体征变化、血液流变学两方面指标评价醋香附不同提取部位疏肝解郁化瘀的药效学差异性,筛选出最佳活性部位。结果:给药后,醋香附不同提取部位组大鼠各项指标均有所改善。体表瘀斑消散、乙酸乙酯组、乙醚组、正丁醇组能显著性降低全血切变率1的水平,乙酸乙酯组亦能显著性减低全血切变率5的水平(与模型组比较,P<0.05);石油醚组、乙醚组、正丁醇组可显著性降低血浆粘度及红细胞压积水平(与模型组比较,P<0.05);乙酸乙酯组、水提组只能降低红细胞压积水平(与模型组比较,P<0.05);正丁醇组、水提组可显著性降低血沉方程K值和红细胞聚集指数(与模型组比较,P<0.05);石油醚组能显著性升高全血高切还原粘度(与模型组比较,P<0.05);石油醚组、乙醚组、乙酸乙酯组、正丁醇组、水提组可显著性减低血浆纤维蛋白原水平(与模型组比较,P<0.05))。结论:乙酸乙酯部位及石油醚部位为醋香附疏肝散瘀的主要有效提取部位,初步认为醋香附是通过降低血液中的血浆纤维蛋白原、降低血液黏度以达到活血化瘀的功效。
Objective: Based on traditional Chinese medicine, rat model of liver disease with liver stagnation and blood stasis was established to explore the material basis for effective treatment of liver stagnation and blood stasis of Xiangfu prepared with vinegar, and screen for the effective parts. Method: The model was established using random stimulating methods and epinephrine hydrochloride. The indexes of sign characteristics, blood theology were used to evaluate the pharmacodynamics diversity of different extracts of Xiangfu prepared with vinegar and select the best active parts. Result: The indicators were improved after drug administration including dissipating of the surface ecchymosis. Ethyl acetate group, ether group and n-butanol group significantly decreased the level of blood shear rate 1 compare to model group (P〈0.05). Ethyl acetate group also significantly reduced the level of blood shear rate 5 (P〈0.05). Petroleum ether group, ether group, n-butanol group significantly decreased the level of plasma viscosity and hematokrit level (P〈0.05). Ethyl acetate group, water extract group only reduced the level of hematokrit (P〈0.05). N-butanol group, water extract group significantly reduced K value of erythrocyte sedimentation rate and red cell assembling index (P〈0.05). Petroleum ether group significantly increased whole blood higher shear reductive viscosity (P〈0.05). Petroleum ether group, ether group, ethyl acetate group, n-butanol group, water extract group significantly reduced the level of plasma fibrinogen(P〈0.05). Conclusion: The best active parts were ethyl acetate extract and petroleum ether. It suggests that Xiangfu prepared with vinegar plays a role in promoting blood circulation to remove blood stasis by decreasing plasma fibrinogen and blood viscosity in the blood.
