摘要
目的:建立葎草茎中木樨草素的定量检测HPLC方法,考察葎草茎的木樨草素含量。方法:高效液相色谱法中固定相为Kromasil C_(18)分析柱(200×4.6 mm,5 m),流动相为甲醇∶0.2%磷酸水溶液(55∶45),检测波长为350 nm,流速为1 mL/min,柱温为30℃。结果:木樨草素在0.03~0.46μg范围内浓度与峰面积呈良好线性关系。线性方程为Y=2 607 187X-19 694,r=0.999 4,平均回收率为98.8%,RSD=1.77%(n=5)。结论:所建立的方法简便,准确,专属性强,灵敏度高,重现性好,可用于草中木樨草素的含量测定。
Objective: To establish the quantitative detection methods for determining the contents of luteolin in the stems of humulus japonicus by HPLC, and investigate the contents of luteolin in the stems of humulus japonicus. Methods: Stationary phase of HPLC was Kromasil C18 analytical column (200×4.6 mm, 5 m), mobile phase was: methanol:0.2% aqueous phosphate solution (55:45), detection wavelength was 350 rim, flow rate was 1 mL/min, column temperature:30℃. Results: The concentrations and peak area of luteolin were in good linear relationship in the range from 0.03 to 0.46 μg. Linear equation was: Y=2 607 187X-19 694, r=0.999 4, average recovery rate was 98.8%, RSD=1.77%(n=5). Conclusion: The method, simple, accurate, with high specificity, high sensibility and good repeatability, could be used for content determination ofluteolin in the stems ofhumulus japonicus. humulus japonicus; luteolin; HPLC
