摘要
目的寻找一种更加简便的方法来定性检测流感病毒抗原阳性细胞。方法设计流式的方法来检测甲型流感病毒(IA)和乙型流感病毒(IB)抗原阳性细胞。采用2种荧光素标记抗体进行流式标记鼻咽拭子的上皮细胞,对157份鼻咽拭子进行甲型和乙型流感病毒抗原检测。结果 157例样本中,直接免疫荧光法检测到甲型流感病毒阳性44例,乙型流感病毒阳性35例,流式法检测到甲型流感病毒阳性39例,乙型流感病毒阳性29例,两种方法检测甲型和乙型流感病毒的阳性率差异无统计学意义(P>0.05)。两种方法检测IA的阳性与阴性总符合率为93.0%,阳性率差异无统计学意义(P>0.05),一致性系数检验κ=0.820,提示两种检测方法一致性较强且有统计学意义(P<0.05)。两种方法检测IB的阳性与阴性总符合率89.8%,阳性率差异无统计学意义(P>0.05),一致性系数检验κ=0.687,两种检测方法一致性基本满意且有统计学意义(P<0.05)。结论采用流式法可及早快速筛查甲型、乙型流感标本,操作步骤简便,只需不到2 h就可完成检测,可以推广应用。
Objective The domestic current mainmethods to detect influenza virus A and B antigens positive cells is direct immunofluorescence,which is troublesome. The aim of this study was to find a simple qualitative method for the detection of influenza virus A and B antigens positive cells. Methods Two specific antibodies labelled with different fluorescein were used for flow cytometry. A total of 157 nasopharyngeal swab specimens were collected for the antigen detection of influenza viruses A and B by flow cytometry method. Results Direct immunofluorescence method showed that 44 cases were positive for influenza A virus,and 35 cases were positive for influenza B virus. Flow cytometry method indicated that influenza A virus was found in 39 cases,and influenza B virus in 29 cases. There were no obvious differences in direct immunofluorescence detection and flow cytometry detection tested by McNemar′s test(P〉0.05). Conclusion Flow cytometryis a rapid and easy antigen-detecting assay for influenza virusesand can be used for screening and diagnosis of influenza A and B virus.
