摘要
目的:阐明山楂叶活性成分牡荆素-4″-O-葡萄糖苷在大鼠体内药物特征。方法:采用反相高效液相色谱法进行测定,牡荆素-4″-O-葡萄糖苷多剂量及多途径给药后大鼠血浆中含量。结果:静脉给药牡荆素-4″-O-葡萄糖苷与口服相比,静脉给药牡荆素-4″-O-葡萄糖苷在大鼠血浆中药-时曲线的符合三室模型,口服给药牡荆素-4″-O-葡萄糖苷在大鼠血浆中药-时曲线的符合二室模型。在大鼠体内360 min内基本消除,符合非线性动力学过程行为。结论:实验首次建立了HPLC法研究山楂叶单体成分牡荆素-4″-O-葡萄糖苷在大鼠体内药代动力学。
Objective:To study the pharmacokinetics of vitexin-4″-O-glycoside in hawthorn leaf in rats to provide a reliable analysis method.Methods:By using HPLC technology to research pharmacokinetics characteristics of vitexin-4″-O-glycoside after administration in vein and oral administration at three single doses in rats.Results:The intravenous dose of vitexin-4″-O-glycoside was 3-chamber model and the oral administration was 2-chamber model.Within 360 minutes,vitexin-4″-O-glycoside was basically eliminated,conformed to the nonlinear dynamic process.Conclusion:The experiment established an HPLC method for the first time to determine vitexin-4″-O-glycoside in rats’ plasma,with strong specificity,which presents a high sensitivity,fast analysis,good precision and accuracy,and is suitable to the pharmacokinetic study for vitexin-4″-O-glycoside.
