摘要 利用DNA条形码技术对燕窝进行基源鉴别。提取燕窝基因组DNA,选择多细胞动物线粒体细胞色素氧化酶I(CO I)的通用引物进行DNA扩增。PCR产物测序分析后,输入Gen Bank中进行BLAST搜索,分析样品遗传距离、构建NJ系统发育树,同时利用实时荧光PCR法对燕窝中是否掺有猪源性成分进行检测。结果显示,收集的15个样品中,13个样品与爪哇金丝燕Aerodramus fuciphagus的CO I DNA序列相似度高于99%,其余2个样品无法获取目的 PCR产物;实时荧光定量PCR结果显示15个样品的Ct值均大于35,样品检测含猪源成分呈阴性。因此,DNA条形码技术为判断燕窝的基源提供了一个可行方法。 To identify the original species of cubilose by DNA Barcoding technique,Genomic DNA was extracted from cubilose and PCR was performed with the metazoan Mitochondrial Cytochrome Oxidase I(CO I) degenerate primers. The amplified sequence was aligned in the Gen Bank database,the nearest distances were calculated and a NJ-tree was built as well. Real-time PCR was applied to detect whether swine-originated ingredient was mixed in the cubilose or not. The results showed that 13 out of the 15 collected samples,CO I DNA sequence similarities to Aerodramus fuciphagus were higher than 99%,and the target PCR products could not be obtained from the other 2 samples. Results of the Real-time PCR showed that Ct values of the 15 samples were all over 35,swine-originated ingredient was not detected.DNA barcoding is a potential technical tool to identify the original species of the edible cubilose.
