反相高效液相色谱法测定牡丹皮中苯甲酸和苯甲酰基总苷 被引量:1

摘要 目的建立反相高效液相色谱法测定牡丹皮药材中游离苯甲酸和苯甲酰基总苷的含量。方法色谱柱为Agilent Eclipse XDB C_(18)柱(250 mm×4.6 mm,5μm),流动相:乙腈-0.1%磷酸水溶液(25∶75),流速:1.0 m L·min^(-1),柱温35℃,检测波长230 nm;苯甲酰基总苷以总苷碱水解为苯甲酸计。结果游离苯甲酸和苯甲酰基总苷进样量均在0.010 16~0.2032μg与峰面积积分值呈良好的线性关系(r=0.9997),平均回收率分别为99.0%、100.6%,RSD分别为2.6%、2.4%(n=6)。结论该方法简便、准确、重现性好、回收率高,增加苯甲酰基总苷的含量测定可更好地控制牡丹皮药材的质量。 Objective To establish an RP-HPLC method to determine the contents of free benzoic acid and total benzoyl glucosides in Cortex Moutan. Methods The analysis was performed on a Agilent Eclipse XDB C18(250 mm×4.6 mm, 5 μm) column. The mobile phase was acetonitrile and 0.1% phosphoric acid(25∶75) at a flow rate of 1.0 m L·min^-1. The column temperature was 35 ℃ and the detection wavelength was 230 nm. With alkaline hydrolysis, the total benzoyl glucosides in Cortex Moutan were hydrolyzed into benzoic acid, determined by RPHPLC, and calculated as benzoic acid content. Results The linear ranges of free benzoic acid and total benzoyl glucosides were at 0.010 16-0.2032 μg(r = 0.9997). The average recovery was 99.0% and 100.6%; RSD was 2.6% and 2.4%(n = 6). Conclusion This method is simple, accurate, sensitive and reliable, and it can be used for the quality control of Cortex Moutan.
