甘蔗与滇蔗茅杂交后代F_1、BC_1表型和GISH分析 被引量:7

Analysis of Intergeneric Hybrids(F_1, BC_1) of Saccharum×Erianthusrockii using Morphology and GISH Technique

摘要 滇蔗茅是甘蔗重要的近缘植物资源,在甘蔗种质创新和遗传改良中具有重要利用价值。为了鉴定甘蔗与滇蔗茅远缘杂交F_1、BC_1真实性后代种质并探明其遗传物质组成,本研究对远缘杂交组合亲本和后代材料进行了形态学观察、染色体计数、核型分析和染色体荧光原位杂交(genomic in situ hybridization,GISH)分析。结果:(1)表型鉴定结果显示,F1叶片具父本滇蔗茅特征,BC1后代叶片则趋于甘蔗品种。BC1的节间长度和株高表现出一定的杂种优势,茎径和锤度则逐渐趋于甘蔗品种(ROC10);(2)细胞学鉴定结果显示:父本云南95-20 2n=30,核型属2B类型。F1云野03-316 2n=54,属2B类型;3个BC1后代材料2n均为110,其中云野06-278属2B类型,云野06-277和云野06-279属2C类型。(3)GISH结果:在F1和3个BC1材料中均检测到15条滇蔗茅野生种外源染色体,但没有检测到重组染色体。以上结果表明F1和BC1是真实的杂交后代种质,杂交过程染色体遗传方式分别为n+n和2n+n。本研究可为滇蔗茅有利基因向甘蔗转移提供细胞学证据,为滇蔗茅在甘蔗杂交创新中利用方式的制定提供参考。 Erianth us rockii is an im portant, closely related genus of Saccharum L., with a great value in sugarcane innovation and variety improvement. In order to identify the real F1 and BC1hybrids of Saccharum×Erianthus rockii, and prove the genetic composition, the morphological, cytological and GISH(genomic in situ hybridization) technique were used to analyze the parental, F1 and BC1hybrids materials. The results show that:(1) the leaves of F1 progeny were observed alike to the male parent of Erianthus rockii and the leaves of BC1 clones were more alike with sugarcane. The BC1 hybrids appeared heterosis in the internode length and plant height, while the diameter and brix were tended to similarity to sugarcane cultivated variety(ROC10);(2) The cytology identification results has shown that: the somatic chromosome number of the male patent Yunnan 95-20(Erianthus rockii) was 2n=30 with a 2 B karyotype; the F1 clone Yunye 03-316 chromosome number was 2n=54,the karyotype was 2 B; all of the three BC1 hybrids have a same somatic chromosome number 2n =110, but different karyotype: the Yunye 06-278 was 2 B type, and both Yunye 06-277 and Yunye 06-279 were 2 C type;(3)the result of the GISH shown that there were 15 Erianthus rockiiwide chromosomes were detected in F1 and three BC1 hybrids, but not any recombination chromosome was detected by GISH. All the results indicated that the F1 and the three BC1 hybrids were the real progenies of Saccharum×Erianthus rockii intergeneric hybridization, and the chromosome transmission method was n+n and 2n+n respectively. This study can provide the cytological evidence for identification of the advantageous genes of Erianthus rockii transfer to sugarcane, and provide a utilization method reference for using Erianthus rockii in the sugarcane innovation process.
