miR-483-5p抑制BALB/c裸鼠体内胶质瘤生长

摘要 目的研究miR-483-5p在体内对胶质瘤生长的影响。方法建立裸鼠皮下胶质瘤移植瘤动物模型,分为U87细胞对照组及miR-483-5p组,于接种细胞后观测瘤体变化,并于24 d后称量瘤体重量。应用免疫组化方法检测增殖细胞核抗原(PCNA)的表达,观察两组移植瘤细胞增殖变化情况。结果 miR-483-5p抑制胶质瘤移植瘤生长,其生长速度及瘤体重量与对照组比较有统计学差异(P<0.05);miR-483-5p使移植瘤PCNA指数下降,与对照组相比较有统计学差异(P<0.05)。结论 miR-483-5p能够抑制裸鼠脑胶质瘤PCNA表达并抑制瘤体生长。 Objective This study aimed to investigate the anti-glioma activity of miR-483-5p in vivo. Methods U87-control and U87-miR-483-5p cells were respectively and subcutaneously inoculated in one side of flanks of BALB/c nude mice to establish xenograft model of human glioma. The tumor growth of the two groups was observed and examined according to the tumor volume measured termly. Then,the tumors were weighted at the 24 th day after inoculation. Besides,immunohistochemical staining method was used to detect PCNA positive expression rate between the two groups. Results The results showed that miR-483-5p suppressed glioma growth in vivo. Compared with U87-control group,both tumor volume and tumor weight were significantly decreased in U87-miR-483-5p group( P 0. 05). Additionally,PCNA positive expression rate was also decreased in U87-miR-483-5p group( P 0.05). Conclusions miR-483-5p suppressed glioma growth in BALB/c nude mice xenograft model. Low expression of PCNA may serve as one of the mechanism which showed inhibited cellular proliferation in gliomas.
