摘要
为了获得冰灯玉露新品种,采用冰灯玉露松散型胚性愈伤组织为诱变试材,采用浸泡法和在培养基中加入诱变剂的方法进行离体诱变。通过采用不同浓度EMS和不同处理时间,找到愈伤组织半致死剂量的EMS使用浓度和时间,并在此条件下对冰灯玉露松散型胚性愈伤组织进行离体诱变,获得形态变化的突变体。结果显示,采用在培养基中加入0.1%EMS处理4 d可以获得理想的半致死效果。以此条件处理冰灯玉露愈伤组织,通过再生培养可以获得1.5%的形态变异的个体,这些个体的遗传变异情况有待进一步检测。
In order to obtain the new variety of Haworthia cooperivar, the embryogenic callus of Haworthia cooperivar was used as mutagenic material. The mutagenesis in vitro was experimented by immersion method and the method by adding mutagen in medium.By using different concentration of EMS and different treatment time, the median lethal dose was found. The embryogenic callus of Haworthia cooperivar was mutated in vitro by this method, and the mutant of morphological change was obtained. The results showed that the ideal median lethal dose could be obtained by adding 0.1% EMS to the medium and treating for 4 days. By this method, after the callus regenerated, 1.5% of the morphological variation individuals were obtained, but the genetic variation of these individuals need to be further tested.
