莲雾果实高质量总蛋白提取方法的建立

摘要 为了高质量提取莲雾果实总蛋白,选择TCA/丙酮法、匀浆法、酚抽提法、试剂盒法和裂解法,分别从莲雾果实中提取总蛋白,使用Bradford法测定蛋白含量,对蛋白含量较高的匀浆法、酚抽提法和试剂盒法进行SDS-PAGE电泳。结果显示,3种方法所得图谱都条带清晰且较无明显拖尾,其中酚抽提法所得图谱最完整清晰且蛋白得率最高,说明酚抽提法适合莲雾果实总蛋白提取。 In order to establish the method of high quality extraction of total protein of syzygium samarangense,the total protein was extracted from the fruit of rhamnus officinalis by TCA/acetone method,homogenization method,phenol extraction method,kit method and lysis method respectively. Protein content was determined by Bradford method,SDSPAGE was performed on homogenization,phenol extraction and reagent method. The results showed that the patterns of the three methods were clear and had no obvious tailing,and the deduced extraction pattern was the most complete and the highest protein yield.
