摘要
目的探讨淫羊藿苷(ICA)对脂多糖(LPS)诱导的阿尔茨海默病(AD)小鼠模型炎症反应的抑制作用及其学习记忆能力的改善作用。方法卵巢切除ICR小鼠18只,随机分为对照组、LPS组、LPS+ICA组,各6只。对照组灌胃给予二甲基亚砜,LPS组小鼠侧脑室注射LPS建立小鼠AD炎症模型并灌胃给予二甲基亚砜,LPS+ICA组小鼠侧脑室注射LPS后连续灌胃给予ICA,连续用药12d。应用水迷宫实验检测小鼠的学习记忆能力,实时反转录聚合酶链式反应方法检测诱导型一氧化氮合酶(iNOS)和肿瘤坏死因子-α(TNF-α)基因的表达情况。结果与对照组比较,侧脑室注射LPS可显著降低小鼠目的象限时间所占比例和平台穿越次数,显著上调小鼠海马区iNOS和TNF-α基因的表达;而灌胃给予ICA可明显对抗LPS的损伤作用,提高小鼠目的象限时间所占比例和平台穿越次数,降低iNOS和TNF-α基因表达,差异有显著性(F=9.214~57.090,q=4.114~13.400,P<0.05)。结论 ICA能够抑制LPS诱导的AD炎症模型小鼠海马iNOS和TNF-α基因的表达,改善小鼠的学习记忆能力。
Objective To investigate the effect of icariin(ICA)in inhibiting inflammatory resoponse and improving learning and memory abilities in ICR mice with lipopolysaccharide(LPS)-induced Alzheimer's disease(AD). Methods A total of 18 ovariectomized mice were randomly divided into control group,LPS group,and ICA+LPS group,with 6 mice in each group.The mice in the control group were given dimethyl sulfoxide by gavage.The mice in the LPS group were given injection of LPS in the lateral ventricle to establish a mouse model of AD,followed by dimethyl sulfoxide by gavage.The mice in the ICA+LPS group were given injection of LPS in the lateral ventricle and then continuously given ICA by gavage.The course of treatment was 12 days.The water maze test was used to test learning and memory abilities,real-time reverse transcription-polymerase chain reaction was used to measure the mRNA expression of inducible nitric oxide synthase(iNOS)and tumor necrosis factor-α(TNF-α). Results Compared with the control group,the LPS group had significant reductions in the proportion of time spent in the target quadrant and number of platform crossings and significant increases in the mRNA expression of iNOSand TNF-αin the hippocampus.Compared with the LPS group,the ICA+LPS group had significantly less injury induced by LPS,significant increases in the proportion of time spent in the target quadrant and number of platform crossings,and significant reductions in the mRNA expression of iNOSand TNF-α(F=9.214-57.090,q=4.114-13.400,P〈0.05). Conclusion In ICR mice with LPS-induced AD and inflammation,ICA can inhibit the mRNA expression of iNOSand TNF-αin the hippocampus and improve their learning and memory abilities.
