摘要
合成制备了一种荧光标记试剂(2-(9吖啶酮)-乙酰肼),并成功应用于高压液相色谱法测定脂肪酸。2-(9吖啶酮)-乙酰肼与脂肪酸的衍生反应可在20分钟内完成。衍生物具有良好的荧光性质,激发和发射波长分别为260 nm和430nm。采用梯度洗脱使得20种脂肪酸在BDS–C8反相柱上得到良好分离,20种脂肪酸的线性系数均>0.996。当采用20mg栝楼样品进行分析时,脂肪酸检出限(三倍信噪比)在0.21–1.20μg/g之间。采用这种荧光试剂衍生的方法对栝楼根、茎、叶和果皮中的游离脂肪酸的含量进行定量分析。
A new labeling reagent,2-( 9-oxoacridin-10( 9H)-yl) acetohydrazide( OAAH),has been synthesized and successfully applied to the HPLC determination of fatty acids of Trichosanthes kirilowii Maxim. The reaction of OAAH with fatty acids proceeded quickly in the presence of 1-ethyl-3-( 3-dimethylaminopropyl)-carbodiimide hydrochloride( EDC) as condensing reagent within 20 min. The derivatives exhibit excellent fluorescence property with excitation and emission wavelengths of 260 nm and 430 nm,respectively. The 20 derivatives of fatty acids were separated on a BDS C8 reversed-phase column with gradient elution. Good linear correlations were observed for all fatty acids with correlation coefficients of >0.996. When 20 mg of sample was used for analysis,the detection limits at a signal-to-noise ratio of 3 were in the range of 0.21 – 1.20 μg/g. Free fatty acids in the roots,stems,leaves and peels of Trichosanthes kirilowii Maxim were analyzed by the developed method.
