锌硒茶对壬基酚致大鼠代谢综合症的保护作用研究 被引量:1

The protective effects of zinc selenium tea on rats in metabolic syndrome induced by nonylphenol

摘要 目的:探讨锌硒茶是否对壬基酚暴露致大鼠代谢综合症有保护作用。方法:30只健康SD大鼠[(150±10)g]随机分为对照组(C)、NP组(NP)、NP+锌硒茶组(NP+Zn Se),剂量分别为对照组(玉米油),NP组(NP200μg/kg·d),NP+锌硒茶组(NP200μg/kg·d+锌硒茶1.20 g/kg·d)。每日清晨8点对大鼠进行空腹灌胃NP,自由摄食,下午4点灌胃锌硒茶,灌胃体积均为10 mL/kg·d。每30天监测体重、血糖和血脂,连续灌胃90 d,麻醉后剖杀大鼠,腹主动脉取全血,离心取血清,ELISA试剂盒检测胰岛素、瘦素水平,全自动生化分析仪检测天冬氨酸丙氨酸氨基转移酶(ALT)和天冬氨酸氨基转移酶(AST);取脂肪(性腺旁脂肪垫)、胰腺、肝脏等组织称重后计算脏器系数;分别取适量组织于10%甲醛中固定后HE染色观察病理改变。结果:与对照组相比,NP组及NP+锌硒茶组胰腺系数降低(p<0.05);NP组大鼠90 d体重、血糖、瘦素、脂肪系数、肝脏系数升高(p<0.05),NP组大鼠ALT、AST 2项指标均值均高于对照组和NP+锌硒茶组,但差异无统计学意义。胰腺、肝脏组织出现严重细胞水肿、坏死,胰腺组织出现炎症细胞浸润等病理改变。NP+锌硒茶组血糖、瘦素、脂肪系数较对照组升高,均低于NP组(p<0.05),胰腺系数低于对照组(p<0.05),胰腺、肝脏组织均出现细胞水肿、坏死等病理变化,但程度较NP暴露组较轻。结论:凤冈锌硒茶可拮抗NP致代谢综合症毒作用。 Objective: This study was designed to investigate whether zinc selenium tea showed the protective effects on rats in metabolic syndrome induced by nonylphenol. Methods: The 30 healthy Sprague Dawley rats were divided into randomly three groups, control group(C) with corn oil, nonylphenol group(NP) with NP 200 μg/kg·d, nonylphenol and zinc selenium group(ZnSe- T) with NP 200 μg/kg·d+ zinc selenium tea 1.20 g/kg·d. The rats were given gavages of corn oil or NP with 10 mL/kg·d starting at 8 o'clock a.m and zinc selenium tea at 4 o'clock p.m. Body weight, blood glucose(FBG), triglyceride(TG) were measured monthly. Hepatic tissue, pancreatic tissue and adipose tissues were weighed and organ coefficient were calculated when they were killed on 90 th day. The blood samples of the rats were collected and analyzed for FBG, TG, alanine transaminase(ALT), aspartate transaminase(AST), leptin(LEP) and insulin(INS). The tissues were fixed in a paraformaldehyde solution for 24 h and embedded in paraffin, the tissues were taken histologic examination. Result: The pancreas coefficient of NP group and NP+ zinc selenium tea group were higher than control group(p〈0.05).The weight, blood glucose, leptin levels, adipose coefficient, liver coefficient of NP group increased(p〈0.05). Liver and pancreatic tissues hematoxylin-eosin staining showed necrotic cells, cellular edema, tissue raritas in NP group. Even in part of pancreatic tissues, they showed inflammatory cell infiltration. The levels of FBG, leptip, adipose coefficient on NP+ ZnSe- T group were higher than control group(p〈0.05), while the levels above of blood and panceas confficients were lower than NP group(p〈0.05). Compared with NP group, pathological changes degree of liver and pancreas of NP+ ZnSe- T group were more gentiy. Conclusions: Zinc selenium tea showed the protective effects on rats in metabolic syndrome induced by nonylphenol.
