摘要
目的采用幼龄大鼠慢性腹泻模型,评价小儿止泻膏的止泻作用,并探讨其作用机制。方法采用ig番泻叶制备幼龄大鼠慢性腹泻模型,给予蒙脱石散1.62 g/kg和小儿止泻膏低、中、高剂量(2.03、4.05、8.10 g/kg)进行治疗,给药1、3 d,分别测定稀便率、稀便级、腹泻指数;给药第4天,测定小肠含水量并取血,比色法测定血清中琥珀酸脱氢酶(SDH)、淀粉酶活力、D-木糖,酶联免疫法测定血清D-乳酸、白介素-1β(IL-1β)及肿瘤坏死因子-α(TNF-α)水平。结果给药1次,小儿止泻膏4.05、8.10 g/kg剂量组均可明显降低稀便率,8.10 g/kg剂量组明显降低腹泻指数;给药3次,小儿止泻膏2.03、4.05、8.10 g/kg剂量组均能明显降低稀便率,4.05、8.10 g/kg剂量组明显降低腹泻指数,降低血清D-乳酸水平,8.10 g/kg剂量组能明显升高血清D-木糖水平。小儿止泻膏对SDH、淀粉酶活力及IL-1β、TNF-α水平均无明显影响。结论小儿止泻膏对幼龄大鼠慢性腹泻有明显的治疗作用,主要作用机制是改善肠道的吸收功能及通透性。
Objective To evaluate the therapeutic effect of Xiao＆#39;er Zhixie Paste （XZP） by using the young rat model of chronic diarrhea,and to explore its mechanism.Methods Chronic diarrhea model in young rats was induced by ig senna.Rats were ig with Montmorillonite powder of 1.62 g/kg,XZP of low,medium,and high dose （2.03,4.05,and 8.10 g/kg） for treatment.Loose stools rate,loose stool grade and diarrhea index were determined 1 and 3 d after treatment respectively.The water content of small intestine was measured and blood was collected for testing serum succinate dehydrogenase （SDH）,amylase,D-xylose by colorimetric determination,testing serum D-lactic acid,IL-1 β,and TNF-α by Elisa after administration.Results The rate of loose stools in XZP 4.05 and 8.10 g/kg dose group,and diarrhea index in 8.10 g/kg dose group significantly reduced after the first treatment.The loose stools rate of XZP 2.03,4.05,and 8.10 g/kg dose group,diarrhea index,serum D-lactic acid level in 4.05,8.10 g/kg group significantly reduced,and serum D-xylose level in 8.10 g/kg dose group significantly increased 3 d after treatment.However,XZP had no significant effect on SDH,amylase activity and IL-1β,TNF-α levels.Conclusion XZP has obvious therapeutic effect on chronic diarrhea in young rats,the mechanism is to increase improve the absorptive function and permeability of intestinal tract.
