PPARγ在AMPK激活调节心肌肥厚中的作用

摘要 目的:探讨过氧化物酶体增殖物激活受体(PPAR)γ是否参与单磷酸腺苷激活的蛋白激酶(AMPK)激活对心肌肥厚调节。方法:采用[3H]亮氯酸掺入、RT-PCR和Western blot等实验方法,观察AMPK激活与抑制时对笨肾上腺素(PE)诱导的培养新生SD大鼠心肌细胞肥大的影响及PPARγ的mRNA与蛋白表述的变化。结果:AMPK激活或抑制可抑制或增加PE诱导心肌细胞蛋白合成,同时伴随有PPARγ mRNA及蛋白表达的增减。结论:PPARγ参与了AMPK激活对心肌肥厚的调节。 Objective :To investigate that peroxisome proliferator activated Teceptor (PPAR) gamma is involved in the regulation of cardiac hypertrophy by activation of adenosine monophosphate activated protein kinase (AMPK).Method: To understand the effect of hypertrophy and expressions of mRNA and protein of PPAR gamma in the cultured myocardial cells of newborn SD rats induced by phenylephrine (PE)and AMPK activation, through [3H]-leucine incorporation, RT-PCR and Western blot.Results: AMPK activation or inhibition can inhibit or increase myocardial cell protein synthesis induced by PE, accompanied by the increase of mRNA and protein expression of PPAR gamma. Conclusion: PPAR γ is involved in the regulation of cardiac hypertrophy by AMPK activation.
