红景天苷对乳鼠心肌肥大细胞的保护作用研究

摘要 目的在原代培养的新生大鼠心肌细胞上,观察红景天苷(sal)对苯肾上腺素(PE)诱导的心肌细胞肥大的影响。方法应用细胞培养技术培养新生大鼠心肌细胞,采用倒置显微镜和图像分析测量细胞面积,测定心肌细胞。H.亮氨酸掺入率作为心肌细胞肥大的指标;用Western blot法检测心肌细胞热休克蛋HSP70蛋白的表达。结果浓度50Hg/ml的sal能抑制PE诱导的心肌细胞增大及心肌细胞合成速率的增加(P〈0.05);能显著增加心肌肥大细胞HSP70蛋白表达(P〈0.05)。结论sal可以抑制PE诱导的心肌细胞肥大,这可能与其增加HsP70蛋白表达有关。 Objective To explore the regulatory effect of salidroside ( Sal ) on HSP70 expression in improving rat myocardial hypertrophy. Methods Cultured cardiomyocytes Of nude rats were used to establish a myocardial hypertrophy model by phenylephrine ( PE ) stimulation. The cardiomyocyte surface area was detected by inverted microscope and digital image analysis technology. 3H-leucine incorporation was measured to evaluate myocardial hypertrophy. The expression of HSP70 protein was detected by Western blot. Results Sal ( 50p.g/ml ) was able to decrease the increased cell surface area and the 3H-leucine incorporation was stimulated by PE ( P〈0.05 ) . Sal was able to increase HSP70 protein expression. Conclusion Sal can ameliorate myocardial hypertrophy, probably through increases HSP70 protein expression.
