香菇杂交菌株酯酶同工酶分析 被引量:3

摘要 [目的]采用酯酶同工酶技术对原生质体单核化对称杂交获得的后代及其亲本进行遗传分析。[方法]研究以香菇菌株168、931和野生香菇及其杂交后代为材料,采用酯酶同工酶技术,研究香菇原生质体杂交后代与亲本之间的遗传关系。[结果]供试菌株酯酶同工酶酶谱带数为4~9条,迁移率在0.092~0.738之间。所有杂交后代与亲本之间既有共同酶带,又有属于自己的特征酶带,可初步认定供试的10个杂交后代有别于亲本,与前期的拮抗试验结果相符。在相异系数为0.60时,以59和xy2为亲本的杂交群体分为三类,第一类为59,第二类为xy2,第三类为ln3、ln4和ln8;在相异系数为0.70时,以ztd和xy2为亲本的杂交群体分为两类,野生菌株xy2单独为一类。[结论]聚类分析结果初步鉴定了杂交亲本与后代之间的亲缘关系,为以后栽培实验提供理论依据。 [Objective] The aim was to find the genetic relationship between parents and generations with esterase isozyme. [Method] Shiitake strains 168, 931. wild Shitaka species and their hybrids were used as materials, the genetic relationship between parents and filial generations gained by protoplast hybridization technology were studied by using esterase isoenzyme technic. [Result] The results showed that the esterase isoenzyme zymogram bands fur all strains ranged from 4 to 9, the migration rate was between 0.092 and 0.738. There were common enzyme bands between hybrids and parents, and all the off-springs had their own characteristics bands, which indicated that the selected 10 hybrids were different from the parent, the same with earlier antagonistic test results. When the dil'ferent eoeffi^-ient is 0.60, clustering analysis resuhs showed that the population derived fi'om parents 59 and xy2 was dMded into three groups, the first group included only 59, the second group only xy2, the third group, three strains, In3, In4 anti ln8; when the different coefficient is 0.70, the population derived ['tom parents ztd and xy2 was divided into two groups, wild strains xv2 was separated ahme in lhe first group anti all the others in the second group, indicating xy2 has a distant genetic' distance with olher strains. [Conclusion[ Clnster analysis resuhs preliminarily identified the genetic relationship between parents and hybrids, which provide the theoretical basis for future cultivation experiment.
