Simultaneous Determination of 2,3,5,4'-Tetrahydroxy stilbene-2-Ο-β-D-glucoside,Salvianolic Acid B,and Puerarin in Jiangzhi Huoxue Tablet by HPLC

摘要 目的:建立同时测定降脂活血片中活性成分2,3,5,4-四羟基二苯乙烯-2-O-β-D-葡萄糖苷(二苯乙烯苷)、丹酚酸B、葛根素含量的HPLC方法。方法:色谱柱为Diamonsil C18(250 mm×4.6 mm,5μm),柱温为35℃,流速为1.0 mL·min^(-1),进样体积为10μL,检测波长为286 nm,以0.1%磷酸水为流动相A,以乙腈为流动相B。结果:二苯乙烯苷的质量浓度在2.674~342.3μg·mL^(-1)(r=0.9994)的范围内、丹酚酸B的质量浓度在2.731~349.3μg·mL^(-1)(r=0.9994)的范围内、葛根素的质量浓度在2.857~365.7μg·mL^(-1)(r=0.9995)的范围内与峰面积线性关系良好,精密度RSD值均小于5%。结论:经方法学验证,该法能同时测定3种成分的含量,可以满足降脂活血片的质量测定要求。 Objective To establish a method for simultaneous determination of three constituents 2,3,5,4′-Tetrahydroxy stilbene-2-O-β-D-glucoside, Salvianolic acid B, and puerarin in Jiangzhi Huoxue Tablet. Methods Jiangzhi Huoxue Tablet was dissolved in methanol, 10 μL solution was sampled. HPLC was adopted to determine the concentration of the three constituents using Diamonsil C18 chromatography column (250 mm×4.6 mm, 5 μm) at 35 ℃. The components, flow rate of mobile phase, and detection wavelength were optimized. Results The three components were successfully determined simultaneously using HPLC under the flow rate 1.0 mL· min-1 and detection wavelength 286 nm with the mobile phase 0. 1% phosphoric acid water (A)-acetonitrile (B). The linear ranges for determination of 2,3,5,4′-Tetrahydroxy stilbene -2-O-β-D-glucoside, Salvianolic acid B, and Puerarin were 2.674~342.3 μg · mL-l(r= 0.9994), 2.731~ 349.3 μg · mL-l(r=0. 9994), 2.857-365.7 μg · mL-1(r=0.9995), respectively, with RSD 〈5%. Conclusion A simple and accurate HPLC method has been established to simultaneously determine 2,3,5,4′-Tetrahydroxy stilbene-2-O-β-D-gluco- side, Salvianolic acid B, and puerarin in Jiangzhi Huoxue Tablet.
