摘要 目的:研究前列腺癌组织中miR-21和程序性死亡因子-4(PDCD4)mRNA的表达水平。方法:临床纳入我院2013年10月至2016年4月期间行手术治疗的26例前列腺癌患者,术中分别取患者前列腺癌组织及癌旁正常前列腺组织各26对。采用荧光定量PCR对所取组织进行检测,检测所取标本中miR-21以及PDCD4 mRNA表达情况,采用Western印迹检测前列腺癌及癌旁组织中PDCD4蛋白的表达水平,分析miR-21与PDCD4表达水平的关系。结果:前列腺癌中miR-21表达水平相对于癌旁组织来说更高(P<0.05);而PDCD4 mRNA表达水平明显低于癌旁组织(P<0.05)。癌旁组织PDCD4蛋白表达灰度值比值明显高于前列腺癌(P<0.05)。前列腺癌miR-21与PDCD4 mRNA表达水平呈显著负相关(P<0.05);与PDCD4蛋白表达水平也呈明显负相关(P<0.05)。结论:miR-21可能通过下调PDCD4表达水平从而参与前列腺癌的发病机制,从而引起前列腺癌的发生和发展。 Objectives: To study the expression of miR-21 and programmed death factor-4(PDCD4)mRNA in prostate cancer tissue. Methods: 26 prostate cancer patients in our hospital from October 2013 to April2016 were selected. The prostate carcinoma tissue and normal adjacent tissues of the 26 patients were taken and detected by fluorescence quantitative PCR,to determine the expression of miR-21 and PDCD4 mRNA specimens.The expression of PDCD 4 protein in prostate cancer and adjacent tissues was detected by Western blot detection and the correlation between miR-21 and the expression of PDCD4 was analyzed. Results: The expression level of miR-21 in prostate cancer tissue was higher than that in adjacent tissue(P〈0. 05),while the expression level of PDCD4 mRNA was significantly lower than that of adjacent tissues(P〈0. 05). The expression of PDCD4 protein in carcinoma gray value ratio was significantly higher than that of prostate cancer tissue(P〈0. 05). There was a significant negative correlation between miR-21 and PDCD4 mRNA expression in prostate cancer tissue(P〈0. 05),and the expression level was negatively correlated with PDCD4(P〈0. 05). Conclusion: The miR-21 may be involved in the pathogenesis of prostate cancer by down regulating the expression level of PDCD4,which can lead to the occurrence and development of prostate cancer.
