东北地区珍贵阔叶树种裂叶榆离体快繁初步研究 被引量:1

The Preliminary Study on Rapid Propagation in Vitro of Rare Broad-leaved Species Ulmus laciniata in Northeast Region

摘要 为建立裂叶榆组织培养体系,选取裂叶榆木质化和水培萌发幼嫩茎段进行离体培养,探讨不同消毒时间、激素种类和浓度对裂叶榆离体快繁的影响。表明:木质化茎段在0.1%升汞消毒14 min时,污染率稍低,褐化率低,成活率高;幼嫩茎段在0.1%升汞消毒3 min时幼嫩茎段污染率最低,褐化率较低;在对休眠芽和幼嫩茎段进行离体培养时筛选出MS+2.0 mg/L 6-BA+0.1 mg/L NAA为最佳培养基激素组合,且在BA:NAA浓度比为20:1时,培养15 d后,裂叶榆的腋芽萌发率最高(80%),而且萌发芽的腋芽绿色,长势良好。 In order to establish a tissue culture system of Ulmus laciniata, taking lignified and hydroponic budding tender stem segments in U. laciniata as materials, researching the effects of different disinfection time, media types and hormone concentrations on propagation in vitro of U. laciniata. The results showed that when lignified stem segments were taken materials by use 0.1% HgC12 for 14 min, the pollution rate was slightly lower, browning rate was slow and survival rate was high. When tender stem segments were taken materials by use 0.1% HgC12 for 3 min, the pollution rate was the lowest, browning rate was lower. The optimal medium and hormone combination was MS+2.0 mg/L 6-BA+0.1 mg/L when dormant buds and tender stem segments were taken materials. Also when the concentration ratio was 20:1 for BA and NAA, after fifteen days, the germination rate of axillary buds was up to 80% for U. laeiniata, the axillary bud was green and grew well.
