摘要
阿尔茨海默症(Alzheimer’s disease,AD)是以老年斑、神经元纤维缠结和神经元丢失为主要病理改变的进行性神经变性疾病。随着全球老年化速度的加快,老年痴呆症对家庭和社会造成重大影响。海洋蕴藏丰富的生物资源,海洋环境的特殊性为人们提供了寻找特异性、高活性功效因子的可能。食源性硫酸多糖作为广受关注的功效因子之一,早在抗血栓、抗病毒等方面表现出突出的作用。近年来研究发现其在抗AD方面表现出非凡的作用。相比于目前临床治疗AD的药物存在疗效不确切或毒副作用大,口服吸收差的缺点,食源性硫酸多糖及其低分子质量衍生物具有特效,低毒,易透过血脑屏障等先天优势。更重要的是,这些食源性硫酸多糖类在海洋动植物食品中广泛存在,通过日常饮食摄入即可获得,无需昂贵的药物代价,因而研究食源性海洋硫酸多糖的神经保护构效机理具有现实意义。本文系统综述AD的发病机理以及食源性海洋硫酸多糖抗老年痴呆的作用机制,以期为食源性海洋硫酸多糖抗老年痴呆的功效提供有效依据。
Alzheimer's disease (AD) is a chronic, progressive, neurodegenerative disorder of the brain, and has been characterised by the presence of amyloid plaques (APs), neurofibrillary tangles (NFTs) and Neuronal loss. With the acceleration of the global aging, AD has brought a heavy economic burden to both families and the society. The ma- rine environment is a rich source of biocompounds with significant and specific biological activities, sulfated polysaccha- rides is one of spieces which have been the most wildly researched. It has been reported to have anticoagulant, anti- thrombotic and anti-virus activities, et.al. Recently, numerous reports have also found it show a promising potential in anti-AD. Compared with the current clinical drugs for AD, which exist the disadvantage of inaccuracy, toxic side effects and poor absorption, marine sulfated polysaccharide and its low molecular weight derivatives showed better potential in specific, low toxicity and the easy access to the blood-brain barrier, et al. More importantly, these marine sulfated polysaccharides are widely spread in marine flora and fauna food, people can easily intake by daily dietary without the cost of expensive drugs. Therefore, the study on neuroprotective mechanism of sulfated polysaccharide from marine food has practical significance. In the current review, we summarize the pathogenesis of AD as well as neuroprotective mech- anism of food-derived marine sulfated polysaccharide, which lays an thesis basis on the developing marine functional food for anti-Alzheimer.
