琼脂寡糖的制备及其对免疫细胞的作用 被引量:2

摘要 琼脂糖是一种生物惰性材料,可降解成小分子量寡糖来提高其生物活性。由盐酸降解和葡聚糖凝胶柱分离纯化得到琼脂寡糖。通过红外光谱和飞行质谱对琼脂寡糖进行结构分析和分子量测定。通过细胞增殖、细胞形貌观察和荧光定量PCR实验研究琼脂寡糖对巨噬细胞的作用。结果表明琼脂寡糖作用于RAW264.7巨噬细胞没有明显的细胞毒性作用,细胞形态完整。琼脂寡糖明显抑制LPS刺激下巨噬细胞的IL-1β,IL-6和TNF-α的mRNA表达;在无LPS诱导作用下,琼脂寡糖也不刺激巨噬细胞合成促炎因子。研究初步表明琼脂寡糖有望作为一种无免疫刺激活性的生物材料。 Agarose is a kind of bio-inert material and its biological activity may be enhanced by degrading into agaro-oligosaccharides with low molecular weight. Agaro-oligosaccharides were obtained from acid degradation of agarose and purified by glucan gel column.The structures and molecular weight of the agarooligosaccharides were analyzed by FT-IR and MALDI-TOF-MS.The effect of the agaro-oligosaccharides on macrophages was detected by cell proliferation,cell morphology and qPCR.Results showed that the agarooligosaccharides had no significant cytotoxicity on RAW264.7 macrophages,and their cell morphology was intact.The agaro-oligosaccharides obviously inhibited the mRNA expression levels of IL-1β,IL-6and TNF-αof the LPS-induced macrophages, and had no situmaltion on proinflammatory cytokines expression of macrophages.These demonstrate that agaro-oligosaccharide is potentially a biomaterial without immune stimulating activity.
