石榴皮对非酒精性脂肪肝病的基因表达调控数据分析 被引量:4

摘要 目的:应用生物信息学分析方法,阐述石榴皮对非酒精性脂肪肝病(NAFLD)的基因表达调控机制。方法:运用生物信息学分析工具QOE、DAVID和STRING,对NCBI的GEO数据库中石榴皮对NAFLD可能治疗作用的相关数据进行挖掘分析。结果:分析喂养与未喂养石榴皮的高脂饮食大鼠基因芯片数据,筛选出8个差异表达基因。其中包括7个高表达基因:CAD、C1QBP、RPL10、RPS7、NHP2、CARM1、LPL;1个低表达基因:NR3C1。这些基因主要涉及的功能包括:嘧啶核酸的生物合成、RNA的修复与加工、核糖体的生物合成、核糖核蛋白复合体、细胞质和细胞器重要物质、脂蛋白生物功能相关基因等。结论:石榴皮可能通过上调LPL、CARM1、C1QBP、NHP2、RPL10、RPS7、CAD和下调NR3C1基因表达的分子机制,促进甘油三酯(TG)的水解,降低血液中游离脂肪酸的含量,缓解胰岛素抵抗,增强肝细胞的抗氧化应激能力及维持线粒体功能的稳定,进而维持肝细胞的正常生物代谢,从而对NAFLD具有一定的疗效。 Objective:To select the different genes expression regulation of rats with non-alcoholic fatty liver disease which were treated with pomegranate peel versus by applying boinformatics analysis tools, and to analyze the possible function of these different ex- pression genes in disease. Methods :The bioinformatics analysis tools as QOE, DAVID and STRING were used for analysis ,with the data downloaded from the GEO database of NCBI. Results : Eight different expressed genes were selected,including seven significantly highly expressed genes as CAD, C I QBP, RPL10, RPST, NHP2, CARM t, LPL ; and one lowly expressed gene NR3 C I. There were some main bi- ological function of this eight different expressed genes,which including the biosynthesis of pyrimidine nucleotide, the repairing and pro- cessing of RNA, the biosynthesis of ribosome and ribonucleoprotein complex, the important matter of cytoplasm and organelles,the rele- vant genes of lipoprotein biological function, etc. Conclusion : The results shows that the treatment of pomegranate peel for non-alcoholic fatty liver disease can up-regulate the expression of LPL, CARM1, C1QBP, NHP2, RPL10, RPST, CAD and down-regulate the expression of NR3C1, to promote the hydrolysis of triglycerides, and pomegranate peel can reduce the content of free fatty acid in the blood, and re- lieve the insulin resistance, strengthen liver cell oxidative stress resistance and maintain the stability of the mitochondrial function, and maintain the normal metabolism of hepatocytes to guarantee the pomegranate peel the non-alcoholic fatty liver disease.
