摘要
目的研究青蒿琥酯抑制胃腺癌细胞生长及调控细胞周期作用。方法利用流式细胞术方法检测不同胃病变组织中细胞周期素D1(CyclinD1)蛋白表达水平。不同浓度(0、30、60、120μmol/L)青蒿琥酯(artesunate,Art)干预胃腺癌SGC-7901细胞24h,0μmol/L Art采用生理盐水代替药物作为对照组,流式细胞术检测细胞周期及CyclinD1蛋白表达水平,并分析SGC-7901细胞中CyclinD1蛋白表达水平与细胞周期G0/1期相关性。结果胃腺癌组织中CyclinD1蛋白表达水平显著高于正常胃组织中的表达水平,P<0.01。不同浓度Art组与对照组相比,SGC-7901细胞周期G0/1期显著增多,P<0.01,且具有Art剂量依赖性,而CyclinD1蛋白表达水平显著降低,P<0.01,具有Art剂量依赖性;CyclinD1蛋白表达与细胞周期G0/1期呈显著负相关,P<0.01。结论 CyclinD1在胃腺癌组织中的高表达导致了细胞周期紊乱,从而参与了胃腺癌发生及发展,Art诱导胃腺癌SGC-7901细胞周期阻滞于G0/1期,与下调细胞中CyclinD1蛋白表达有关,从而起到抗胃腺癌作用。
Objective To study the inhibitory effect of artesunate on the growth and cell cycle of gastric adenocarcinoma cells.Methods To detect the Cyclin D1 protein expression in different tissues of gastric disease, cellcycle and Cyclin D1 protein expression in SGC- 7901 cells were detected by flow cytometry after the treat- ment of different concentrations of artesunate (0,30,60,120 μmol/L) for 24h,0 μmol/L Art with saline instead as control group. The correlationship between G0/1 phase of SGC - 7901 cells and Cyclin D1 protein expression in SGC- 7901 cells was analyzed by SPSS software. Results Cyclin D1 protein expression level in gastric adenocarcinoma was significantly higher than that in normal gastric tissues, P 〈0.01. Compared with the control group, the G0/1 phase of SGC - 7901 cells was significantly increased (P 〈0.01 ) in different concentrations of Art groups,in an Art dose dependent manner. Compared with the control group,the Cyclin D1 protein expression of SGC-7901 cells was significantly decreased (P 〈0.01) in Art groups,in an Art dose dependent manner. The Cyclin D1 protein expression in SGC- 7901 cells was negatively correlated with the G0/1 phase of SGC- 7901 cells,P〈0.01. Conclusion The abnormal high expression of Cyclin D1 results in disorder of cell cycle,and it participates in the occurrence and development of gastric adenocarcinoma. Art induces gastric adenocarcinoma SGC- 7901 cell cycle arrest in the G0/1 phase, which is related to the down regulation of Cyclin D1 protein expression,which may play an important role in the inhibition of gastric adenocarcinoma.
